摘要
目的:研究α-硫辛酸对糖尿病肾病(DN)自噬相关因子LC3、Rab7和Beclin1表达的影响。方法:2型糖尿病肾病Ⅲ期患者110例,其中55例常规治疗为对照组,55例患者在常规治疗的基础上加用硫辛酸治疗为研究组;建立DN模型大鼠,对比硫辛酸治疗的DN患者和DN模型组大鼠Scr、BUN水平和肾脏组织中LC3、Rab7和Beclin1的表达的影响。结果:治疗后两组患者Scr和BUN均显著降低(P<0.05),且研究组患者治疗后Scr和BUN均显著低于对照组患者(P<0.05),肾脏组织中LC3、Rab7和Beclin1蛋白表达均显著高于对照组(P<0.05);DN模型大鼠经硫辛酸治疗后,Scr和BUN均显著降低(P<0.05),肾脏组织中LC3、Rab7和Beclin1基因表达显著升高(P<0.05)。结论:α-硫辛酸对糖尿病肾病肾脏的保护作用可能通过促进肾脏中自噬相关因子LC3、Rab7和Beclin1的表达有关。
Objective: To study the effects of α-lipoic acid on expression of autophagy related factors LC3,Rab7 and Beclin1 in diabetic nephropathy. Methods: There were 110 cases of patients which were phase Ⅲ type 2 diabetic nephropathy,55 patients underwent routine treatment as the control group,and others were treated with lipoic acid on the basis of conventional therapy as research group.The BUN and Scr before and after treatment and the expression of LC3,Rab7 and Beclin1 proteins in renal tissue after treatment in two group of patients and three groups of rats were compared. Results: There was no significant difference in BUN and Scr between the two groups of patients before treatment,but they all dropped significantly after treament,and the research group decreased more significantly than control group ( P <0.05).After treament,the expression of LC3,Rab7 and Beclin1 proteins in research group was significantly higher than control group ( P <0.05).The BUN and Scr in DN rats had significantly went down and the expression of LC3,Rab7 and Beclin1 had significantly increased after treatment with lipoic acid. Conclusion: The protective effect of α-lipoic acid on the kidney of diabetic nephropathy may be related to the expression of autophagy-related factors LC3,Rab7 and Beclin1 in the kidney.
