葡萄糖浓度及α-硫辛酸对培养的内皮细胞NF-κB表达的影响 被引量:1

摘要 目的观察葡萄糖浓度对培养的内皮细胞NF-κB表达的影响及α-硫辛酸的作用。方法培养人脐静脉内皮细胞,先分为高糖干预24h(H24组)和72h(H72组),设正常糖干预24h(N24组)和72h(N72组)为对照,之后将细胞由高糖环境转至正常糖环境24h(H72-N24组)或者同时给予α-硫辛酸(H72-Nα24组),设一直用正常糖干预为对照。共聚焦显微镜观察细胞胞浆及胞核的荧光,测定不同时间点NF-кB表达、总SOD活性和MDA含量。结果 (1)H24组与N24组相比,三项指标差异均无显著性。H72组与N72组及H24组相比,三项指标差异均有显著性,其中NF-кB表达量、MDA含量高于N72组(均P<0.01)及H24组(均P<0.05);SOD活性低于N72组(P<0.01)及H24组(P<0.05)。(2)H72-N24组与H72-Nα24组及N72-N24组相比,三项指标差异均有显著性,其中NF-кB表达量、MDA含量高(均P<0.01),SOD活性低(均P<0.01)。H72-Nα24组与N72-N24组相比,三项指标差异均无显著性。结论 (1)高糖通过氧化应激促进内皮细胞NF-кB表达有时间依赖性;(2)从高糖环境转至正常糖环境短时间内存在炎症记忆作用;(3)纠正高糖并加用α-硫辛酸可以解除炎症记忆作用。
