摘要
目的:探索脊髓背角胶状质(substantia gelatinosa,SG)神经元去极化反跳的电生理特性,进一步阐明疼痛的脊髓调控机理,为慢性疼痛疾病的治疗药物研发提供科学依据。方法:取4周龄雄性SD大鼠66只,随机分为3组(n=22):模型组(坐骨神经部分结扎,PSNL)、假手术组(sham)和对照组(control)。测量三组大鼠机械痛阈,膜片钳记录去极化反跳等电生理特性,进行对比分析。结果:(1)模型组术后7~14d机械痛阈明显低于另外两组(P<0.05);(2)每组SG神经元均有50%以上出现去极化反跳现象,而且模型组去极化反跳出现的概率明显高于另外两组(P<0.05);(3)模型组SG神经元的动作电位阈值明显低于其他两组,输入阻抗和动作电位频率均高于另外两组(P<0.05)。结论:大鼠坐骨神经部分结扎术后去极化反跳现象增加,神经元兴奋性增高,出现机械痛敏,提示SG神经元去极化反跳可能参与慢性疼痛的调控。
Objective:In order to explore the characteristics of rebound depolarization of substantia gelatinosa (SG) neurons of spinal cord in neuropathic pain rats.Methods:4-week-old male Sprague-Dawley rats were randomly divided into three groups (n = 22):partial sciatic nerve ligation (PSNL) group,sham operation group and control group.The mechanical threshold of the 3 groups was tested,and the rebound depolarization and other basic electrophysiology characteristics were recorded by whole cell patch clamp, then we compared the data of the 3 groups.Results:(1) The mechanical threshold of the PSNL group on 7-14 days after operation was significantly lower than the other two groups (P < 0.05);(2) More than 50% of the SG neurons in each group showed rebound depolarization,and the probability of rebound depolarization in the PSNL group was significantly higher than the other two groups (P < 0.05);(3) The action potential threshold was lower and the input resistance (Rin) and the action potential frequency were higher in the PSNL group compared with the other two groups (P < 0.05).Conclusion:After PSNL surgery,the occurrence probability of rebound depolarization and neuronal excitability increase,and mechanical hyperalgesia appears,which suggest that rebound depolarization of SG neurons may be involved in the modulation of chronic pain.
