HIV感染者外周血PD-1、TIGIT基因表达水平与T细胞分化、凋亡分子分泌的相关性 被引量:2

The correlation between the gene expression of PD-1 and TIGIT in peripheral blood and T cell differentiation and apoptotic molecule secretion in HIV-infected patients

摘要 目的分析人类免疫缺陷病毒(HIV)感染者外周血程序性死亡分子1(PD-1)、T细胞免疫球蛋白及免疫酪氨酸样抑制基序(TIGIT)表达水平与T细胞分化、凋亡分子分泌的相关性。方法 选择2013年6月—2018年8月四川省甘孜藏族自治州人民医院传染病科确诊为HIV感染的患者80例作为HIV组,同期于医院体检的健康者60例作为健康对照组。采集2组外周血后测定PD-1、TIGIT基因表达水平、T细胞亚群的的数目、凋亡分子的含量。结果 与健康对照组比较,HIV组外周血中PD-L1及TIGITmRNA表达水平、Th2、Th9、Treg细胞的数目以及血清Fas、FasL、TNF-α、TRAIL的含量均明显升高(t/P=22.399/0.000、17.504/0.000、19.838/0.000、34.614/0.000、18.963/0.000、26.822/0.000、14.005/0.000、21.289/0.000、21.987/0.000),外周血Th1、Th17细胞的数目以及血清Bcl-2的含量均明显降低(t/P=27.206/0.000、16.579/0.000、23.095/0.000);HIV组外周血PD-L1及TIGITmRNA表达水平与Th1、Th17细胞的数目以及Bcl-2的含量呈负相关(r=-0.623、-0.591、-0.657及-0.452、-0.524、-0.612,P均<0.05),与Th2、Th9、Treg细胞的数目以及FasL、Fas、TNF-α、TRAIL的含量呈正相关(r=0.475、0.563、0.608、0.498、0.632、0.574、0.538及0.475、0.551、0.561、0.494、0.601、0.526、0.583,P均<0.05)。结论 HIV感染者外周血PD-1、TIGIT的高表达能够影响CD4+T细胞亚群的分化以及凋亡分子的分泌。 Objective To analyze the correlation between the expression levels of programmed death molecule 1 (PD-1), T cell immunoglobulin and immunotyrosine-like inhibitory motif (TIGIT) in peripheral blood of HIV infected patients and T cell differentiation and apoptotic molecule secretion. Methods From June 2013 to August 2018, 80 patients diagnosed as HIV infection in the Infectious Diseases Department of the People's Hospital of Ganzi Tibetan Autonomous Prefecture in Sichuan Province were selected as the HIV group, and 60 healthy people in the same period were selected as the healthy control group. The expression of PD-1 and TIGIT, the number of T lymphocyte subsets and the content of apoptotic molecules were measured after collecting peripheral blood from two groups. Results Compared with the healthy control group, the mRNA expression levels of PD-L1 and TIGIT, the number of Th2, Th9, Treg cells and the levels of serum Fas, FasL, TNF-α and TRAIL in the peripheral blood of the HIV group were significantly increased ( P <0.01), the number of Th1 and Th17 cells in peripheral blood and the content of serum Bcl-2 were significantly decreased ( P <0.01);The mRNA expression levels of PD-L1 and TIGIT in peripheral blood of HIV group were negatively correlated with the number of Th1, Th17 cells and Bcl-2 ( r =-0.623 , r =-0.591 , r =-0.657, r =-0.452, r =-0.524, r =-0.612, P <0.05), positively correlated with the number of Th2, Th9, Treg cells and the content of FasL, Fas, TNF-α, TRAIL ( r =0.475, r =0.563, r =0.608, r =0.498, r =0.632, r =0.574, r =0.538, r =0.475, r =0.551, r =0.561, r =0.494, r =0.601, r =0.526, r =0.583, P < 0.05 ). Conclusion The high expression of PD-1 and TIGIT in peripheral blood of HIV-infected patients can affect the differentiation of CD4 +T cell subsets and the secretion of apoptotic molecules.
