杜蛭丸灌胃对缺血性脑卒中大鼠的保护作用及其机制探讨 被引量:4

摘要 目的评价杜蛭丸灌胃对缺血性脑卒中大鼠的保护作用,探讨其可能的作用机制。方法将142只健康雄性SD大鼠随机分为7组:A组(正常对照组,16只,给予等量氯化钠溶液灌胃)和B组[6只,给予中剂量杜蛭丸溶液,1.0g/(kg·d)灌胃];C组、D组、E组和F组各24只,依次分别予等量氯化钠溶液、低剂量杜蛭丸溶液[0.5g/(kg·d)]、中剂量杜蛭丸溶液[1.0g/(kg·d)]、高剂量杜蛭丸溶液[2.0g/(kg·d)]灌胃;G组(阳性对照组,24只)第1~7天不予任何处理,第8天时在大鼠尾静脉注射依诺肝素钠(500U/kg),除G组外余各组均1次/d,连续7d进行药物干预。第8天除A组和B组外,余各组均采用线栓法建立大鼠大脑中动脉阻塞(MCAO)模型,缺血再灌注24h后,评价各组大鼠神经行为学评分和脑梗死百分率,检测大鼠血小板聚集率、血细胞比容、凝血和血液黏度等。结果本研究共有102只大鼠造模成功。各组大鼠神经行为学评分和脑梗死百分率比较差异均有统计学意义(P<0.05),其中F组和G组大鼠神经行为学评分、脑梗死百分率低于C组(P<0.01)。体外实验时,血小板同腺苷二磷酸在不同浓度的杜蛭丸水溶液(0.1mg/mL,0.3mg/mL和1.0mg/mL)中充分反应时,随着杜蛭丸溶液浓度的逐渐升高,血小板聚集率逐渐下降(均P<0.01)。各组MCAO模型大鼠血细胞比容比较差异有统计学意义(P<0.01),其中C组大鼠血细胞比容明显高于A组(P<0.01),G组、F组大鼠血细胞比容明显低于C组(P<0.01)。各组大鼠低切和高切全血黏度比较差异有统计学意义(P<0.05),其中B组低于A组(P<0.05),C组高于A组(P<0.05);各组大鼠低切全血相对黏度比较差异有统计学意义(P<0.05),其中D组、E组和G组低于C组(P<0.05)。各组大鼠凝血酶时间(TT)、纤维蛋白原(FIB)水平比较差异有统计学意义(均P<0.01),其中B组和C组TT、FIB水平高于A组(P<0.05)。结论高剂量杜蛭丸溶液灌胃对MCAO模型大鼠缺血性脑卒中具有保护作用,可� Objective To evaluate the effects of Duzhi pill by oral gavage(o.g.)on ischemic stroke rats and to study the possible mechanisms.Methods A total of 142 male SD rats were randomly divided into 7 groups:group A(control group,n=16,equal volume of saline),group B[medium-dose Duzhi pill solution,n=6,1.0 g/(kg·d)];group C,group D,group E and group F(n=24 respectively)were administered(o.g.)the same amount of saline,low-dose Duzhi pill solution[0.5 g/(kg·d)],medium-dose Duzhi pill solution[1.0 g/(kg·d)],high-dose Duzhi pill solution[2.0 g/(kg·d)];group G(positive control group,n=24)without any treatment from the 1st to 7th day,then received enoxaparin sodium(500 U/kg)via the tail vein on the 8th day.Every group was administered(o.g.)once a day for 7 days except group G.On the 8th day,all groups received the middle cerebral artery occlusion(MCAO)by suture method with the exception of group A and B.After 24 h post-ischemia/reperfusion,the neurobehavioral scores and the percentage of cerebral infarction were evaluated and the platelet aggregation,hematocrit,coagulation and blood viscosity were tested.Results In this study,102 rats were successfully established.There were significant differences in neurobehavioral score and cerebral infarction percentage between each group(P<0.05).The neurobehavioral score and cerebral infarction percentage in group F and G were lower than those in group C(P<0.01).In vitro,when platelets reacted fully with adenosine diphosphate in different concentrations of Duzhi pill aqueous solution(0.1 mg/mL,0.3 mg/mL and 1.0 mg/mL),the platelet aggregation rate decreased gradually with the increase of the concentration of Duzhi pill solution(all P<0.01).Hematocrits of different MCAO model rat groups were statistically significantly different(P<0.01),group C significantly higher than group A(P<0.01),group G and group F significantly lower than group C(P<0.01).There were significant differences between low-shear and high-shear whole blood viscosity in each group(P<0.05),group B lower than group A(P<0
