摘要
目的探究微小RNA-150-5p(miR-150-5p)在冠心病发生发展中的作用。方法利用miRDB数据库进行miR-150-5p的靶基因预测,将预测结果导入DAVID在线工具进行靶基因KEGG功能富集分析。将SD大鼠分为实验组和对照组,每组各12只,分别以高脂饮食喂养和冠脉接扎法建立冠心病大鼠模型。同时对空白组以假手术处理。实验组静脉注射miR-150-5p类似物,连续3 d给药后使用WB检测wnt信号通路相关蛋白GSK-3α、p-GSK-3α、GSK-3β、p-GSK-3β及P53表达量。对照组静脉注射300μg对照miRNA类似物,给药时间、检测时间和检测指标同实验组。结果 miR-150-5p调控的靶基因集与Wnt信号通路、癌症发生、内分泌调节等分子途径和细胞行为相关。miR-150-5p对GSK-3α、p-GSK-3α、GSK-3β、p-GSK-3β的表达量几乎没有影响。与空白组及对照组相比,miR-150-5p明显下调实验组p53的蛋白表达量。结论 miR-150-5p通过抑制下游靶基因P53来调控Wnt信号通路参与细胞增殖与凋亡,可能为诱发冠心病的潜在机制。
Objective To explore the function of microRNA-150-5 p in the occurrence and development of coronary heart disease. Methods The target genes of microRNA-150-5 p were predicted by using the microRNA DB database, and the predicted results were imported into DAVID online tool for functional enrichment analysis of target gene KEGG. The model animals(rats) were divided into experimental group and control group with 12 rats in each group. The rat model of coronary heart disease was established by feeding high fat diet and ligating coronary artery. At the same time, the blank group was treated with sham operation. microRNA-150-5 p analogues were injected intravenously in the experimental group and detected after three consecutive days of administration. The control group received intravenous injection of 300 μg control microRNAs analogues. Results Bioinformatics analysis showed that the target gene set regulated by microRNA-150-5 p was related to Wnt signaling pathway, cancer occurrence, endocrine regulation and cell behavior. The expression levels of GSK-3 a, p-GSK-3 a, GSK-3 beta and p-GSK-3 beta were hardly affected by microRNA-150-5 p. Compared with the blank group and the control group, the expression of P53 protein in the experimental group was significantly down-regulated by microRNA-150-5 p. Conclusion MicroRNA-150-5 p regulates Wnt signaling pathway through inhibiting downstream target gene P53 to participate in cell proliferation and apoptosis, which is considered as a potential target to induce coronary heart disease.
