吸烟、赖氨酰氧化酶与非小细胞肺癌发生发展的相关性 被引量:4

摘要 目的研究赖氨酰氧化酶(LOX)在吸烟与非吸烟患者非小细胞肺癌组织、癌旁组织、正常肺组织中的表达,探讨LOX与非小细胞肺癌发生发展的关系,吸烟与LOX的关系,吸烟与非小细胞肺癌的关系。方法实时荧光逆转录聚合酶链式反应(RT-PCR)检测44例非小细胞肺癌组织,癌旁组织,正常肺组织中LOX mRNA含量;Western Blot检测44非小细胞肺癌组织,癌旁组织,正常肺组织中LOX蛋白表达水平。并进行统计学分析。结果所检测组织标本中LOX mRNA及LOX蛋白的表达率均呈阶梯式表现,即非小细胞肺癌组织>癌旁组织>正常肺组织,表达差异有统计学意义(P<0.05)。吸烟组LOX mRNA和LOX蛋白的表达率低于非吸烟组,差异有统计学意义(P<0.05)。结论 44例非小细胞肺癌组织中LOX均呈过度表达状态,LOX可能与非小细胞肺癌的发生发展有关。而吸烟的非小细胞肺癌组织中LOX表达低于非吸烟非小细胞肺癌组织。吸烟可能通过抑制LOX的表达从而影响非小细胞肺癌的发生与发展。 Objective To explore the relationship among smoking, LOX and nonsmall cell lung cancer development through testing the expression of LOX in cancer tissue, adjacent tissue and normal lung tissue for smoking and no- smoking patients with nonsmall cell lung. Methods Realtime fluorescence reverse transcription polymerase chain reaction (RT - PCR) was used to detect LOX mRNA content of 44 cases of nonsmall cell lung cancer and adjacent tissues and normal lung tissue, and LOX protein expression of those samples were detected by western blot method. Results The detection of tissue specimens expression of LOX mRNA and LOX protein expression showed a ladder, that was, the highest in nonsmall cell lung cancer tissue, next in adjacent tissues, the lowestein normal lung tissue, with significantly difference (P〈0.05). Expression of LOX mRNA and LOX protein in smokers group was lower than nosmokers, the difference was statistically significant(P〈0. 05). Conclusions LOX shows overexpression in nonsmall cell lung cancer, it may be associated with the development of nonsmall cell lung cancer. And expression of LOX in smokers was lower than that in n smokers, so smoking may affect the occurrence and development of non- small cell lung cancer by inhibiting the expression of LOX.
