摘要
目的探讨逍遥散加味方对过敏性结膜炎辅助性T细胞1(Th1)/辅助性T细胞2(Th2)平衡的影响。方法采用卵清蛋白(OVA)诱导小鼠过敏性结膜炎模型,随机分为模型组、逍遥散加味方治疗组(包括高、中、低剂量组)、阳性药物对照组(色甘酸钠滴眼液),另设正常组作为空白对照,每组10只,共60只,干预治疗2周。观察小鼠眼部体征、应用酶联免疫吸附法(ELISA)检测血清γ-干扰素(IFN-γ)、白介素4(IL-4)水平。结果(1)各剂量逍遥散加味方均能减轻小鼠眼部症状体征,模型组的体征评分均值最高,明显高于空白对照组(P<0.01),逍遥散加味方各剂量组的体征评分均较模型组降低(P<0.01),各逍遥散加味方组体征评分均值差异不显著,并且与色甘酸钠滴眼液组结果接近(P>0.05)。(2)逍遥散加味方高、中、低剂量组及色甘酸钠滴眼液组的血清IL-4、IL-4/IFN-γ均低于模型组(P<0.01),IFN-γ高于模型组(P<0.01),逍遥散加味方高、中、低剂量组间差异无统计学意义(P>0.05)。结论逍遥散加味方能缓解过敏性结膜炎小鼠的眼部体征,并可能调节Th1/Th2平衡的影响。
Objective To investigate the effect of modified Xiaoyao San on the balance of helper T cell 1( Th1) and helper T cell 2 ( Th2) on allergic conjunctivitis. Methods Mice model with allergic conjunctivitis induced by ovalbumin( OVA) were randomly divided into model group,modified Xiaoyao San group( including high,medium and low dose group),positive drug control group( cromolyn sodium eye drops),another normal group as a blank control,each group had 10 mice so a total of 60,and intervention was given for 2 weeks. Eye signs were observed and serum levels of IFN-γ and IL-4 detected by ELISA. Results Each dose of modified Xiaoyao San could alleviate eye symptoms. The model group had the highest mean sign score,significantly higher than that in the blank control group( P < 0. 01),the scores of signs in Xiaoyao San group with different dose were lower than of in the model group( P < 0. 01). There was no significant difference between the mean scores of Xiaoyao San group and the scores were closer to that of the Cromolyn sodium eye drops group( P > 0. 05). 2. Serum IL-4,IL-4 / IFN-γcontent in each dose of Xiaoyao San group and cromolyn sodium eye drops group were lower than that of model group( P < 0. 01),but IFN-γwere higher than that of model group;there was no significant difference among high,medium and low dose of Xiaoyao San groups( P > 0. 05). Conclusion Modified Xiaoyao San may ease the eye signs in mice with allergic conjunctivitis,and possibly regulate Th1 and Th2 balance.
