摘要
观察平喘灵冲剂对哮喘模型大鼠IgE,白介素-4(IL-4)和γ-干扰素(IFN-γ)表达的影响,探讨该中药治疗哮喘的作用机理。将60只SD大鼠分为6组:正常对照组、模型组、平喘灵大、小剂量组、地塞米松组及平喘灵冲剂小剂量加地塞米松组。采用免疫放射法观察IgE的变化,半定量逆转录-聚合酶链反应(RT-PCR)观察IL-4I、FN-γmRNA的表达。模型组IgE,IL-4 mRNA表达显著升高(P<0.05)I、FN-γmRNA表达显著降低(P<0.05);各治疗组IgE,lL-4mRNA的表达均显著降低(P<0.05)I、FN-γmRNA的表达明显增高(P<0.05);平喘灵冲剂小剂量加地塞米松组作用更明显(P<0.05)。平喘灵冲剂可能是通过减少IL-4的分泌合成,相对增加IFN-γ的合成,使IL-4和IFN-γ之间趋于平衡,从而抑制体内IgE的产生,发挥其抗气道炎症的作用,平喘灵冲剂与地塞米松合用有协同作用。
To investgate the effects of pingchuanling (PCL) on IgE, IL-4rnRNA and IFN-γmRNA in asthmatic rots and the mechanism of PCL in treating asthma. Sixty SD rats were randomly divided into control group, asthmatic group, two PCL-treated groups, dexamethasone-treated group and dexamethasone plusPCL-treatedgroup. Each immuno-radiometricassay(IRMA)and semi-quantitatively reversed transecription-poly merase chain reaction (RT-PCR) were used in this study. Compared to the control group, the asthmatic group had significant increases of serum IgE and IL-4mRNA expression, and a decrease of IFN-γmRNA expression( P 〈 0.05). Compared to the asthmatic group, the PCL-treated groups showed significant reductions of serum IgE and IL-4mRNA expression, and escalation of IFN-γmRNA expression( P 〈 0.05). Conclusion:PCL relieved bronchial asthma by modulating IL-4/IFN-γration followed by IgE level and was synergistic with dexamethasone.
