摘要
目的 动态观察用人蛔虫变应原激发豚鼠哮喘后 ,支气管肺泡灌洗液 (BALF)中炎性细胞总数及细胞分类的动态变化 ,探讨蛔虫变应原致喘机理。方法 用人蛔虫变应原激发豚鼠哮喘后 1h、2 4h、72h ,观察BALF炎性细胞总数及细胞分类的动态变化。结果 在激发后 1h、2 4h、72h阳性激发组豚鼠BALF中白细胞总数分别为 72 8± 6 3,10 0 2± 78,12 74± 12 2 (单位为 :细胞数 /ml) ,远高于阴性对照组 (分别为 4 84± 4 4 ,4 6 6± 6 1,4 98± 73)和佐剂对照组 (分别为 4 88± 75 ,5 13± 10 5 ,4 93± 78) ,有显著性差异 (P <0 .0 1)。阳性激发组中BALF内嗜酸性粒细胞 (EOS)分别为 176± 32 ,30 6± 4 7,2 76±4 9 ,均比正常阴性对照组和佐剂对照组增多 ,具有显著性意义 (P <0 .0 1)。而阳性激发组 72hEOS胞浆中嗜酸性颗粒数目远远低于 2 4h组 ,细胞核浆比增加 ,表明EOS已经大量脱颗粒。结论 用蛔虫变应原诱发豚鼠哮喘后 ,使得大量炎性细胞渗出、浸润 ,对气道上皮形成炎性损伤 。
In order to study the principle of asthma induced by ascaris allergen, the dynamic changes of the total and differential leukocyte counts in bronchoalveolar lavage fluid were examined during ascaris allergen induced asthmatic reaction. Significant increases in the total leukocyte counts in the positive group were observed, compared with those in negative control group and adjuvant control group ( P <0.01). The total eosinophil counts in positive group elevated significantly, compared with those in negative control group and adjuvant control group ( P <0.01). In addition in the positive group, the number of acidophil granule of EOS at 72?h was markedly lower than that at 24?h, with the ratio of nucleus and cytoplasm increasing, suggesting that granules of EOS were released. These findings suggested that challenge with ascaris allergen could induce leukocyte infiltration, which could facilitate the injury of epithelium of airway and bronchial responsiveness.
