摘要
以青龙半木质化枝条为外植体,采用丛生芽诱导途径,建立起组培快繁体系。结果表明:最佳启动培养基为WPM+0.05mg/LTDZ+30g/L蔗糖+6.5g/L琼脂,启动率为96.1%;最佳增殖培养基为WPM+0.06mg/LTDZ+0.3mg/LIBA+6.5g/L琼脂+30g/L蔗糖,继代周期30d,增殖系数高达4.8;最佳生根培养基为1/2WPM+0.3mg/LNAA+6.5g/L琼脂+30g/L蔗糖,培养30d,其生根率达96.7%。炼苗后,移栽到珍珠岩∶蛭石∶泥炭土体积比为1∶1∶1的混合基质中,成活率达到90%以上。
The explants were taken from the late and half lignificational stems with shoot in the adult selected trees. An economic and high effective technology system for rapid propagation of Acer palmatum ‘seiryu’was established, through multiple-shoots proliferation. The result show that the optimal starting medium was modified WPM + 0.05 mg/L TDZ+ 30 g/L sucrose + 6.5 g/L gelatin, the establishment rates were 96.1%;the optimal mulitiplication medium was modified WPM +0.06 mg/L TDZ+0.3 mg/L IBA + 30 g/L sucrose + 6.5 g/L gelatin, the subculture cycle was 30 d, the highest multiplication rate was 4.8;the optimal rooting medium was 1/2 WPM + 0.3 mg/L NAA + 30 g/L sucrose + 6.5 g/L gelatin, the rooting rate has reached 96.7% within 30 d. The transplanting matrix were pearlite,vermiculite and peat (1 : 1 : 1, V / V / V), and its corresponding transplanting survival rate was over 90%.
