孕激素受体在不同发育期牦牛乳腺组织内的分布及表达 被引量:4

Distribution and expression of progesterone receptors in mammary gland of yak during different development stages

摘要 为揭示不同发育期牦牛乳腺组织内孕激素受体(progesterone receptor,PR)的分布及蛋白水平的表达,试验应用SP免疫组织化学方法和Image Pro Plus 6.0分析软件检测不同发育期牦牛乳腺内PR的分布情况,并采用Western-blot技术对乳腺组织内的PR蛋白含量进行半定量测定。结果表明:PR阳性物质主要见于乳腺腺泡上皮细胞、导管上皮细胞、血管内皮细胞及小叶间结缔组织细胞的细胞核中,偶见胞质着色。PR阳性物质平均光密度在整个乳腺发育过程都呈现高-低-高的趋势,青春期PR阳性物质数高于其他几个期,差异显著(P<0.05);妊娠期、泌乳早期、泌乳中期和静止期PR阳性物质数比较差异显著(P<0.05);其中妊娠期和静止期的PR阳性物质数均高于泌乳早期和泌乳中期,但妊娠期和静止期PR阳性物质数差异不显著(P>0.05),泌乳早期和泌乳中期PR阳性物质数差异不显著(P>0.05)。PR阳性物质数的分布表明PR可促进导管分支和腺泡发育。PR蛋白相对表达水平在不同发育期牦牛乳腺组织内明显不同,青春期PR蛋白相对表达水平明显高于其他几个期,泌乳早期和泌乳中期PR蛋白相对表达很少,而静止期开始大量表达。提示在牦牛不同发育期,PR蛋白在乳腺发育及泌乳生物学中可能发挥不同的功能。 In order to reveal the distribution and the expression of progesterone receptors in mammary gland of yak during different developmental periods, the localization and the protein content of PR were tested by using the SP immunohistochemical staining method and Image Pro Plus 6. 0 Analysis Software and the Western-blot method, respectively. The results showed that predominant nuclear staining for PR-positive material were found acinar epithelial cells, ductal epithelial cells, interlobular connective tissue cells and vascular endothelial cells and weak cytoplasmic staining of yak mammary gland during different developmental periods. The average optical density of PR-positive material showed a trend of high-low-high during the development of yak mammary gland, the relative expression of PR-positive material during virgin was significantly higher( P<0. 05) than other stages. Forthermore, the relative expression of PR-positive material during pregnant, early lactating, mid-lactating and non-lactating periods were significantly different (P<0. 05), PR-positive material during pregnant and non-lactating periods were higher than that of early lactating, mid-lactating, while there was no significant difference between the relative expression of PR-positive material during pregnant and non-lactating periods, even if early lactating and mid-lactating period (P>0. 05). The distribution of PR-positive material indicates that PR promotes ductal differentiation and acinar development. The relative expression of PR protein was significantly different during different developmental stages of yak mammary gland tissue. The relative expression of PR protein of yak mammary gland tissue in different developmental stages as measured by western blot was significantly higher than that in other stages. The expression of PR in the early lactating and mid-lactating stages was rarely, while non-lactating period began to express.
