非达霉素产生菌种子工艺研究 被引量:4

摘要 为了提高非达霉素的发酵产量及发酵稳定性,替代种子配方中的动物性原料,对非达霉素产生菌的种子培养温度、配方及工艺进行了研究。结果表明:种子最佳培养温度为30℃,多肽粉和酵母蛋白胨FP102可以替代原种子配方中的牛肉膏和蛋白胨两种动物性氮源。优化后的种子配方为玉米淀粉25g/L,酵母浸粉5g/L,多肽粉3g/L,酵母蛋白胨FP1025g/L,碳酸钙4g/L,pH7.0。确定二级种子工艺为一级种子30℃培养56h,0.1%~0.2%接种量接入二级种子培养基,30℃培养48h接入发酵培养基,在成功去除动物性原料的基础上,非达霉素产量较原种子工艺提高了12.3%。 In order to improve fidaxomicintiter,fermentation stability and replace the animal material in the seed formulation,the seed culture temperature,seed formulation and seed technology of fedamycin-producing strain were studied.The results showed that the optimum temperature of seed culture was 30℃,and the polypeptide powder and yeast peptone FP102 could replace beef paste and peptone,which were the two kinds of animal nitrogen sources of the original seed formulation.The seed formula was determined as follows:corn starch 25 g/L,yeast extract powder 5 g/L,polypeptide powder 3 g/L,yeast peptone FP 102 5 g/L,calcium carbonate 4 g/L,pH 7.0.The second-stage seed technology was determined as follows:the first-stage seeds were cultured at 30℃for 56 h,0.1%-0.2%in the second stage flask,and cultured at 30℃for 48 h in the fermentation medium.On the basis of successful removal of animal raw materials,the titer of fidaxomicin was increased by 12.3%compared with the original condition.
