健脾益肺汤抑制NF-κB/STAT3信号通路改善哮喘模型大鼠气道炎症及气道重塑作用研究 被引量:20

摘要 目的观察健脾益肺汤通过抑制核因子-κB/信号转导及转录激活因子3(NF-κB/STAT3)信号通路改善哮喘模型大鼠气道炎症及气道重塑的作用。方法将30只雄性SD大鼠随机分为对照组、模型组和健脾益肺汤组,采用卵蛋白(OVA)联合氢氧化铝凝胶多次激发致敏法造模,健脾益肺汤组以5 g/(kg·d)进行灌胃,每日1次,持续30 d,对照组和模型组给予等量0.9%氯化钠注射液。末次给药1 h后处死大鼠后进行大鼠肺组织切片观察,测量肺内气管壁厚度和平滑肌厚度。检测肺泡灌洗液中蛋白含量INF-γ、IL-4、IL-6、IL-13含量。应用Western blotting检测大鼠肺组织中NF-κB、p-NF-κB、STAT3和p-STAT3含量。结果与对照组比较,模型组大鼠气管壁厚度和平滑肌厚度增加,肺泡灌洗液中蛋白、IL-4、IL-6和IL-13增加,肺组织中NF-κB、p-NF-κB、STAT3和p-STAT3增加,肺泡灌洗液中IFN-γ降低(P <0.05);给予健脾益肺汤干预后,模型组大鼠气管壁厚度和平滑肌厚度降低,肺泡灌洗液中蛋白、IL-4、IL-6和IL-13降低,肺组织中NF-κB、pNF-κB、STAT3和p-STAT3降低,肺泡灌洗液中IFN-γ增加(P <0.05)。结论健脾益肺汤能改善哮喘模型大鼠气道炎症及气道重塑,其机制可能与抑制NF-κB/STAT3信号通路,减轻大鼠炎性反应有关。 Objective: To investigate the amelioration effect of Jianpi Yifei Decoction on airway inflammation and airway remodeling of asthmatic rat model by inhibiting NF-κB/STAT3 signaling pathway. Methods: 30 male SD rats were randomly divided into the control group,the model group and Jianpi Yifei Decoction group. Asthmatic models were established by OVA challenge and aerosol inhalation. Jianpi Yifei Decoction group was given 5 g/(kg·d) by gavage once a day for 30 days. The other two groups were given the same amount of normal saline. 1 hour after the last administration,the rats were killed;the lung tissue sections were observed;the thickness of trachea wall and smooth muscle were measured,and the related indicators were detected. Results: Compared with the control group,the thickness of tracheal wall and smooth muscle in the model group increased,as well as the levels of protein,IL-4,IL-6 and IL-13 in alveolar lavage fluid,NF-κB,p-NF-κB,STAT3 and p-STAT3 in lung tissue;IFN-γ in alveolar lavage fluid decreased(P < 0.05). After the intervention of Jianpi Yifei Decoction,the thickness of tracheal wall and smooth muscle in the model group decreased,as well as the levels of protein,IL-4,IL-6 and IL-13 in alveolar lavage fluid,NF-κB,p-NF-κB,STAT3 and p-STAT3 in lung tissue;IFN-γ increased in alveolar lavage fluid(P < 0.05). Conclusion: Jianpi Yifei Decoction can ameliorate the airway inflammation and airway remodeling in asthmatic rats. The mechanism may be related to inhibition of NF-κB/STAT3 signaling pathway and alleviation of inflammatory response in rats.
