菊粉对不同时期2型糖尿病小鼠髓源抑制性细胞的影响 被引量:1

Effects of Inulin on Myeloid-derived Suppressor Cells from Diverse Stages of Type 2 Diabetes Mellitus in Mice

摘要 目的探讨菊粉对2型糖尿病髓源抑制性细胞的影响。方法采用4周龄的雌性db/db小鼠作为2型糖尿病模型,共40只。在db/db小鼠第6周龄监测空腹血糖,并行3次糖耐量试验,测空腹血糖<7.0mmol·L^(-1)或葡萄糖负荷后2h血糖在7.8~11.1mmol·L^(-1)为糖尿病前期组(PDM组,n=20),空腹血糖≥11.1mmol·L^(-1)为糖尿病期组(DM组,n=20),再从PDM组和DM组分别随机取出10只作为菊粉干预糖尿病前期组(INU/PDM组)和菊粉干预糖尿病期组(INU/DM组)。采用流式细胞术分别检测各组小鼠外周血、脾脏、肝脏MDSCs及其亚型M-MDSCs的比例及血浆白介素-6(IL-6)、白介素^(-1)0(IL^(-1)0)水平变化。结果与模型对照组(PDM组和DM组)相比,菊粉干预组(INU/PDM组和INU/DM组)小鼠的外周血、脾脏、肝脏的MDSCs及其亚型M-MDSCs均升高(P<0.05);血浆IL-6均减少,血浆IL^(-1)0水平均升高(P均<0.05)。40只小鼠外周血、脾脏及肝脏细胞MDSCs、M-MDSCs与IL-6呈负相关,与IL^(-1)0呈正相关(P<0.01)。结论菊粉干预可增加不同时期T2DM小鼠的MDSCs与M-MDSCs,减少炎症反应,可能成为T2DM治疗的新手段。 Objective To investigate the effects of Inulin on myeloid-derived suppressor cells from diverse stages of type 2 diabetes mellitus(T2DM)in mice.Methods Female db/db mice(4 weeks)were adopted to generate a T2DM model.Fasting blood glucose of each mouse from diverse groups was monitored daily from the week 6 glucose tolerance tests were performed 3 times.Mice with fasting blood glucose<7.0mmol·L^-1,glucose load(2h plasma glucose):7.8-11.1 mmol·L^-1 were defined as the pre-diabetic group(PDM group,n=20).Otherwise,mice with fasting blood glucose≥11.1mmol·L^-1 were defined as the diabetic group(DM group,n=20).Ten mice from the PDM group or the DM group were randomly allocated into inulin intervention of PDM group(INU/PDM group)or inulin intervention of DM group(INU/DM group),respectively.The alterations of the proportion of MDSCs and their subtypes in peripheral blood,spleen and liver from each group were detected by flow cytometry.Results The proportion of MDSCs and M-MDSCs in peripheral blood,spleen and liver were significantly increased in inulin treated groups(INU/PDM group and INU/DM group),compared with that in untreated groups(PDM group and DM group)(P<0.05).Decreased IL-6 and elevated IL-10 in T2DM or PDM were found after inulin treatment(P<0.05).Furthermore,the percentage of MDSCs and M-MDSCs in peripheral blood,spleen and liver were negatively correlated with IL-6,but positively correlated with IL-10(P<0.01),respectively.Conclusion Dietary inulin may significantly increase the proportions of MDSCs and M-MDSCs to suppress inflammatory response in diverse stages of T2DM,which may potentially contribute to a novel strategy for diverse stages of T2DM.
