摘要
目的:探讨长链非编码RNA(long non-coding RNA,lncRNA)牛磺酸上调基因1(taurine up-regulated 1,TUG1)和肺腺癌转移相关转录本1(metastasis associated lung adenocarcinoma transcript 1,MALAT1)在2型糖尿病患者外周血单个核细胞(pe-ripheral blood mononuclear cells,PBMC)中的表达及临床意义。方法:采用实时荧光定量PCR(qRT-PCR)技术检测60例2型糖尿病患者(糖尿病组)及45例健康体检者(健康对照组)PBMC中lncRNA TUG1、MALAT1的表达水平,并分析空腹血糖、糖化血红蛋白及病程对其表达的影响。结果:qRT-PCR检测结果显示,lncRNA TUG1在2型糖尿病患者PBMC中的相对表达水平是健康对照组的6.25倍(P <0.05),lncRNA MALAT1在2型糖尿病患者PBMC中的相对表达水平是健康对照组的3.98倍(P <0.05);在2型糖尿病患者的PBMC中,lncRNA TUG1、MALAT1表达水平随着患者空腹血糖和糖化血红蛋白的升高而增加(P <0.05);同时,lncRNA TUG1、MALAT1表达水平也随着患者病程的延长而增加(P <0.05);ROC曲线示lncRNA TUG1的曲线下面积为0.898(95%CI:0.826~0.970,P <0.001),lncRNA MALAT1的曲线下面积为0.715(95%CI:0.583~0.846,P <0.001)。结论:2型糖尿病患者PBMC中的lncRNA TUG1、MALAT1表达升高,且与血糖水平及病程有关,lncRNA TUG1、MALAT1的表达水平或可作为诊断以及评估2型糖尿病患者病情的生物学标志物。
Objective:The aim of this study was to determine the change of long non-coding RNA(lncRNA)taurine up-regulated 1(TUG1)and metastasis associated lung adenocarcinoma transcript 1(MALAT1)in patients’ peripheral blood mononuclear cells(PBMCs)with T2 DM,expecting to find a biomarker in diagnosing T2 DM. Methods:Real-time quantitative PCR(qRT-PCR)was used to detect the expression levels of lncRNA TUG1 and MALAT1 in 60 patients with type 2 diabetes(the diabetes group)and 45 healthy controls(the healthy control group). The effects of fasting blood glucose,glycated hemoglobin and the course of disease on lncRNA expression were analyzed. Results:The results of qRT-PCR showed that the relative expression level of lncRNA TUG1 in PBMC of T2 DM patients was 6.25 times of the healthy control group(P < 0.05),and the relative expression level of lncRNA MALAT1 in PBMC of T2 MD patients was 3.98 times of the healthy control group(P < 0.05);In PBMC of patients with type 2 diabetes,the expression level of lncRNA MALAT1 and TUG1 rised with the increasing of fasting blood glucose and glycated hemoglobin(P < 0.05);Meanwhile,the expression level of lncRNA TUG1 and MALAT1 also increased with the prolongation of the patient’s disease course(P < 0.05);The ROC curve showed that the area under the curve of lncRNA TUG1 was 0.898(95% confidence interval 0.826-0.970,P < 0.001),and the area under the curve for lncRNA MALAT1 was 0.715(95% confidence interval 0.583-0.846,P < 0.001). Conclusion:The expression of lncRNA TUG1 and MALAT1 in PBMC of patients with T2 MD was elevated,and it was related to blood glucose level and course of disease. The expression level of lncRNA TUG1 and MALAT1 may be used as a biomarker for diagnosis and evaluation of the condition of patients with T2 MD.
