摘要
比较4种不同检验方法对亚利桑那沙门氏菌检测的优缺点,采用实时荧光聚合酶链式反应(polymerase chain reaction,PCR)、VITEK 2全自动微生物鉴定系统、API 20E肠杆菌和其它革兰氏阴性杆菌鉴定系统和国标方法对巧克力中亚利桑那沙门氏菌进行检验。结果表明,在检验结果准确性上VITEK 2全自动微生物鉴定系统对双相亚利桑那亚种沙门氏菌鉴定准确率高达98%,而其他3种方法都不能直接鉴定出准确的菌种类型;在操作步骤和时耗上PCR最佳,其次是VITEK 2全自动微生物鉴定系统。VITEK 2全自动微生物鉴定系统操作简便,检验周期短,且更精准,因此更适合对巧克力中亚利桑那沙门氏菌的检测。
The advantages and disadvantages of four different detection methods for Salmonella arizona were compared. The method of real-time polymerase chain reaction (PCR)、VITEK 2 microbiology identification system、API 20E identification system for Enterobacteriaceae and other non-fastidious gram-negatie rods and national standard were used to detect Salmonella arizona in chocolate.The results showed that the accuracy of VITEK 2 microbiology identification system was 98 % for Salmonella enterica ssp. diarizonae, while the other three methods could not directly identify the exact types of bacteria. PCR was superior to VITEK 2 microbiology identification system in operation steps and time-consuming.VITEK 2 microbiology identification system was easy to operate, short test cycle and more accurate, so it was more suitable for the detection of Salmonella arizona in chocolate.
