沙棘甾醇化合物的分离纯化研究 被引量:5

摘要 以沙棘籽油为研究对象,通过单因素和正交试验对皂化法制备甾醇粗制品的工艺条件进行优化,利用重结晶法和大孔吸附树脂法对甾醇粗制品进行精制和纯化,并用薄层色谱法进行鉴定。结果表明,制备甾醇粗制品的最佳工艺条件为:料液比1∶17.5(g/mL)、溶剂50%KOH∶无水乙醇配比1∶12.5(体积比)、皂化时间1.5 h、皂化温度90℃。在此条件下,甾醇粗制品的得率为2.41%。利用重结晶法对甾醇粗制品进行精制,可得到β-谷甾醇含量为91.41%的甾醇精制品。利用大孔吸附树脂法纯化的结果为:洗脱时间在46 min~60 min内,β-谷甾醇的含量最高,纯度为93.54%,经薄层色谱法鉴定,纯化后的沙棘甾醇为β-谷甾醇。 Taking seed oil of Hippophae rhamnoides L. as the research object, the technological conditions of preparation of sterol crude products by saponification method were optimized by single factor and orthogonal test, and sterol crude products were refined and purified by recrystallization method and macroporous adsorption resin method, and identified by thin layer chromatography. The results showed that the optimum technological conditions of preparation of sterol crude products were that solid-liquid ratio was 1 ∶ 17.5(g/mL), and solvent 50 % KOH ∶ anhydrous ethanol ratio was 1 ∶ 12.5(Volume ratio), and saponification time was 1.5 h, and saponification temperature was 90 ℃. Under these conditions, the yield rate of sterol crude products was 2.41 %. The sterol crude products were refined by recrystallization method, and the sterol refined products containing 91.41 % of β-sitosterol could be obtained. The results of purification by macroporous resin adsorption method showed that the content of β-sitosterol after purification was the highest when the elution time was within 46 min-60 min, which was 93.54 %. The purified sterol of Hippophae rhamnoides L. was β-sitosterol by thin layer chromatography to identity.
