富血小板血浆用于兔半月板损伤修复的研究 被引量:6

摘要 目的分别以制备富血小板血浆凝胶(Platelet-rich plasma gel, PRG)时的不同的激活比例为变量和以富血小板血浆(Platelet-rich plasma, PRP)不同的应用频率为变量,探究二者对兔半月板损伤修复效果的影响。方法建立成年大白兔半月板损伤模型,左右侧分别为实验组和对照组,实验组向关节腔内注射PRP作为干预,对照组无干预。Ⅰ期实验:实验组以富血小板血浆凝胶制备时的激活比例分别为3:1,5:1,7:1,按不同比例分为A,B,C三组,每组18只。在术后第4,8,12周时,每组处死6只取半月板进行大体及组织学观察,分别评价PRP促进半月板损伤修复的能力。Ⅱ期实验:实验组以仅1次、每周1次、每2周1次、每4周1次的干预频率分为A,B,C,D四组,每组12只。在术后第8,12周每组处死6只取半月板进行大体及组织学观察,分别评价PRP促进半月板损伤修复的能力。结果Ⅰ期实验,整理实验数据后对各组愈合结果进行评估,C组优于B组和A组,A,B两组无统计学差异。Ⅱ期实验显示,各组间无统计学差异,但直观看,C组愈合效果似乎优于A,B,D组。结论 7:1比例激活剂激活的富血小板血浆凝胶可能更适合促进半月板白区损伤的修复。在不同的干预频率下,富血小板血浆凝胶促进半月板白区损伤修复的效果无统计学差异,其意义有待进一步研究。 Objective To investigate the effects of different activation ratios of platelet-rich plasma gels(PRG) and variable frequency of platelet-rich plasma(PRP) on repairing effects of rabbit meniscal tears.Methods The adult white rabbit meniscus injury model was established. The left and right sides were the experimental group and the control group. The experimental group was given intraarticular injection of PRP as intervention, and the control group was not interfered. In the first-phase experiment, the activation ratio of the experimental group prepared with platelet-rich plasma gel was 3:1, 5:1, and 7:1,divided into 3 groups with 18 rats in each group. At 4 th, 8 th, and 12 th weeks after surgery, 6 meniscuses were sacrificed in each group for gross and histological observations to assess the ability of PRP to promote meniscal repair. In the second phase of the experiment, the experimental group was divided into 4 groups with 12 animals per group, only once per week, once per week, every 2 weeks, and once every 4 weeks. At the 8 th and 12 th weeks after operation, 6 meniscuses were sacrificed in each group for gross and histological observations to assess the ability of PRP to promote meniscal repair. Results In the first phase of the experiment, the healing results of each group were evaluated after finishing the experimental data. The C group was superior to the B and A groups, and there was no statistical difference between the A and B groups. The second-phase experiment showed that there was no statistical difference between the groups, but intuitively, the healing effect of group C appeared to be better than that of group A, B and D.Conclusion 7:1 Proportionate Activator Activated Platelet-rich plasma gel may be more suitable to promote meniscal white zone damage repair. At different frequencies of intervention, platelet-rich plasma gels did not significantly contribute to the repair of meniscal white areas. The significance of this need to be further studied.
