摘要
本文以甲基营养型芽孢杆菌、多粘类芽胞杆菌、侧砲短芽胞杆菌、解淀粉芽胞杆菌等4种兼性厌氧微生物农药为材料,在不同培养基上用3种不同的培养方式进行芽胞含量检测以探究最佳的检测方法,为微生物农药质量检测提供参考。结果表明,除解淀粉芽砲杆菌外,营养琼脂(Nutrientagar,NA)是其他3种兼性厌氧微生物农药芽胞含量检测的最佳培养基,而解淀粉芽砲杆菌进行芽胞含量检测时推荐使用四号培养基。在最佳培养基培养条件下,对3种培养方式进行比较分析,结果表明对于甲基营养型芽胞杆菌和侧砲短芽胞杆菌,平皿夹层法和倒平皿法有效成分含量检测结果较高,而多粘类芽胞杆菌和解淀粉芽胞杆菌倒平皿法检测结果极显著高于平皿夹层法和涂平皿法(Duncan检验,PwO.01)。
This study focused on four facultative anaerobic microbial pesticides, including Bacillus methylotrophicus, Paenibacilhis polymyxa, Brevibacilhis laterosponis and Bacillus amyloliquefaciens. To develop the most appropriate methods for determining the spore content, three cultural methods with different cultural media were selected, which were aimed to provide reference for quality test of microbial pesticides. The results showed that nutrient agar (NA) was the best medium for the determination of the other 3 microbials than Bacillus amyloliquffaciens, for which No. 4 medium was recommended to be used. Under the optimum culture medium, the three culture methods were compared and analyzed. The results displayed that a higher active ingredient content of Bacillus inethylotrophicus and Brevibacillus lalerosporus was obtained when sandwich plate method and pour plate method were used, and that significantly higher results were obtained for Paenibacillus polymyxa and Bacillus amyloliquefaciens when pour plate method was used compared to plate method and smear plate method.
