摘要
目的探讨三叉神经痛(TN)模型大鼠的疼痛状况与脑内兴奋性氨基酸递质表达的相关性。方法健康SD大鼠32只随机分为对照组16只与模型组16只,模型组于大鼠背部皮下注射硝酸甘油建立TM模型,对照组在大鼠背部皮下注射生理盐水。观察与记录大鼠不同时间段的行为与疼痛状况,采用免疫组化法检测谷氨酸的表达并进行相关性分析。结果模型组大鼠在造模后30 min内都出现挠头、爬笼、耳红等症状,对照组大鼠未出现上述行为变化,表明造模成功。在不同时间段内,模型组的挠头与爬笼次数显著多于对照组(P <0.05)。造模前两组大鼠痛觉反应阈值对比无显著差异(P> 0.05),模型组造模后3~7 d的痛觉反应阈值显著高于对照组(P <0.05)。模型组大鼠三叉神经节的细胞内谷氨酸蛋白的表达高于显著对照组,差异有显著性(P <0.05)。在模型组中,直线相关分析显示造模后7d大鼠的谷氨酸蛋白表达水平与疼痛反应阈值呈正相关性(P <0.05)。结论 TN模型大鼠的疼痛状况与脑内兴奋性氨基酸递质-谷氨酸表达有显著相关性,谷氨酸可能与TN的痛觉传递过程有关。
Objective To explore the correlation between pain and the expression of excitatory amino acid transmitters in trigeminal neuralgia(TN)rats.Methods 32 healthy SD rats were randomly divided into control group(n=16)and model group(n=16).Rats in model group were subcutaneously injected with nitroglycerin to establish TM model,and those in control group were subcutaneously injected with saline.The behavior and pain status of rats were observed and recorded.The expression of glutamate was detected by immunohistochemical method and went into correlation analysis.Results The rats in model group showed head-scratching,cage-climbing and ear redness 30 min after model construction.The control group were without the above signs,implying successful model construction.In different periods of time,the head-scratching cage-climbing frequencies in model group were significantly higher than control group(P<0.05).Before model construction,there was no significant difference in pain threshold between the two groups(P>0.05).The pain threshold 3~7 d after model construction in model group was significantly higher than that in control group(P<0.05).The expression of glutamate in trigeminal ganglion neurons in model group were higher than that in control group(P<0.05).In model group,there were significant positive correlation between the expression level of glutamate and pain response threshold 7 d after model construction in model group(P<0.05).Conclusion There are significant correlation between the pain status and the expression of excitatory amino acid transmitters glutamate in the TN model rats,glutamate may be related to the process of the pain of TN.
