细菌内毒素多次给药对家兔致热灵敏度影响的研究 被引量:2

摘要 目的:探讨细菌内毒素多次给药对家兔致热灵敏度的影响。方法:取符合热原检查法规定的家兔36只随机分成3个组,每组12只。耳缘静脉分别注射细菌内毒素2.5、5、10 EU﹒kg^(-1),连续4次,每次间隔17 d。记录各剂量组家兔每次给药的入选动物数,计算平均升温值、升温率和入选率。结果:2.5、5和10 EU﹒kg^(-1)剂量组之间的平均升温和升温率有明显统计学差异(P<0.05或0.01),呈现剂量依赖性。随着给药次数的增加,除2.5 EU﹒kg^(-1)组的升温率在第2和第3次明显升高外,同一剂量组的平均升温和升温率未产生明显变化,而家兔入选数明显减少。另外,家兔入选率还与细菌内毒素剂量呈明显相关性,剂量越大,入选率越低。结论:连续注射细菌内毒素4次,升温达0.6℃的家兔休息两周以上,其热原灵敏度未明显降低,但随着给药次数的增加其入选率明显降低,且细菌内毒素剂量越高,入选率降低更明显。 Objective:To investigate the rabbit pyrogenicity sensitivity to repeated administration bacterial endotoxin.Method:36 rabbits conforming to pyrogen inspection regulation were divided into 3 groups.The rabbits were injected through ear vein 2.5,5 and 10 EU﹒kg^-1 bacterial endotoxin,respectively,for 4 times with the interval of 17 days.The number of animals per group for each injection was recorded,and the average temperature rise value,temperature rise rate and selection rate were calculated.Result:The average temperature rise value and temperature rise rates among 2.5 EU﹒kg^-1,5 EU﹒kg^-1 and 10 EU﹒kg^-1 groups were significantly different(P<0.05 or 0.01),showing a dose-dependent manner.With the increase of injection times,the average temperature rise value and temperature rise rates of the same dose group did not change obviously except the temperature rise rates of 2.5 EU﹒kg^-1 group significantly increased in the second and third times.But the number of rabbits selected decreased significantly.In addition,the number of rabbits selected was also significantly correlated with the dose of bacterial endotoxin.The larger the dose,the less the number of rabbits was selected.Conclusion:After continuous injection of bacterial endotoxin for 4 times,rabbits whose body temperature rises up to 0.6 ℃ rest for more than two weeks,and their pyrogen sensitivity don’t decrease significantly.However,with the increase of the number of injection,the rate of selection decreases significantly.And the higher the dose of bacterial endotoxin,the decrease in the selection rate is more obvious.
