摘要
目的:探讨电针不同状态大鼠对血清外泌体(Exosomes)表达的影响。方法:正常Wistar大鼠随机分为对照组、电针1d组、电针5d组,每组4只。正常Wistar大鼠右后足足底注射完全弗式佐剂0.1ml/只复制大鼠佐剂性关节炎动物模型,分盐水组、模型、模型针刺组,模型针刺组给予1d、5d、10d、20d四个不同时间点的治疗,每个时间点每组4只。取大鼠腹主动脉血液,离心取血清,采用蛋白免疫印迹法检测不同状态(正常、佐剂性关节炎)大鼠血清中Exosomes的含量,观察针刺对不同状态不同时间点大鼠血清Exosomes表达的影响。结果:发现针刺正常大鼠促有促进血清外泌体的释放的趋势,针刺佐剂性关节炎模型大鼠,在炎症急性期,电针有抑制血清Exosomes的释放的趋势,炎症后期,电针有促进血清Exosomes释放的趋势。结论:针刺不同时期模型大鼠,血清Exosomes表达不同,可能与不同炎症时期外泌体所包含的活性成分不同有关,有待进一步研究。
Objective:To investigate the effect of electroacupuncture on the expression of exosomes in normal and CFA model rats.Methods:Normal Wistar rats were randomly divided into control group,electroacupuncture 1 day group,and electroacupuncture 5 day group,with 4 rats in each group.Normal Wistar rats were injected with complete Freund's adjuvant 0.1 ml/only rat in the right hind paw to copy adjuvant arthritis model.Then rats were divided into saline group,model group,model acupuncture group.The model acupuncture group were treated by four different time points of 1 day,5 days,10 days,and 20 days,with 4 rats in each group at each time point.Abdominal aorta blood of rats was taken,and serum was gotten by centrifugation.To observe the effect of electroacupuncture on the expression of Exosomes,the level of exosomes in the serum of rats in different states(normal,adjuvant arthritis)was determined by Western blotting.Results:It was found that electroacupuncture had a tendency to promote the release of serum-derived exosomes in normal rats,and electroacupuncture RA model rats had a tendency to inhibit the release of serum-derived exosomes in the acute phase of inflammation.In the later stage of inflammation,electroacupuncture had a tendency to promote the release of serum-derived exosomes.Conclusion:The expression of serum-derived exosomes is different in electroacupuncture model rats at different stages.The results might be related to the difference of active substance contained in exosomes at different stages of inflammation,which needs further study.
