植物乳杆菌KF5胞外多糖合成条件的优化研究 被引量:7

Optimization the Synthetic Conditions of Exopolysaccharides Secreted by Lactobacillus plantarum KF5

摘要 从我国西藏传统乳制品发酵剂藏灵菇(Kefir粒)中分离得到一株具有产胞外多糖(exopolysaccharises,EPS)能力的植物乳杆菌菌株KF5,测得其EPS初始产量为49.32 mg/L。通过单因素及正交试验,优化其EPS的最佳合成条件为:培养时间30 h,培养基初始p H 6.3,接种量3%。培养基初始pH值和接种量对EPS的合成影响不显著,培养时间影响显著(p<0.05)。在最优组合条件下,KF5胞外多糖合成量达到95.58 mg/L,相比于正交优化前提高了26.48%。 The Lactobacillus plantarum KF5 was isolated from Kefir grains(Tibet,China),which could secret exopolysaccharises(EPS)during the growing process.The initial yield of EPS was 49.32 mg/L with phenolsulfuric acid method.The conditions for the synthesis of EPS were optimized with single factor and orthogonal experiments.The optimum conditions were as follows:the incubation time 30 h,the initial pH of medium 6.3 and the inoculum volume 3%.Moreover,initial pH of culture medium and inoculum volume had no significant effect on the yield of EPS,but the culture time had significantly influence on the production(p<0.05).The yield of L.plantarum KF5 could reach 95.58 mg/L under the optimum conditions,which was higher more than 26.48%before orthogonal optimization.
