摘要
【目的】系统鉴定玉米纤维素合成酶(CesA)家族成员,明确其进化关系及基因功能。【方法】采用系统生物学方法,在全基因组水平上对玉米CesA家族成员进行鉴定,进一步结合基因结构变化对CesA家族成员的进化关系进行系统阐述;通过RNA测序和核糖体图谱,系统分析玉米CesA家族成员响应干旱胁迫的表达模式,并从基因复制角度分析该家族基因的扩增方式,同时基于构建的转录调控网络,对玉米CesA基因的功能进行初步探索。【结果】鉴定出16个玉米CesA家族成员,分为7个子家族,其中有3个是新发现的成员;CesA家族成员在玉米发育过程中对干旱胁迫的响应有明显的组织特异性和发育时期特异性,干旱胁迫对玉米种子发育过程的影响较对植株生长过程的影响大;CesA家族成员在合成纤维素过程中相互协同作用,成员间存在功能冗余。【结论】系统鉴定出了16个玉米CesA家族成员,进一步明确了其彼此之间的功能协作及其在种子发育和植株生长过程中对干旱胁迫的响应程度。
【Objective】This study identified the CesA family members in maize to clarify the evolution relationship and gene functions.【Method】The system biology methods were used to identify the family members on genome-wide basis.The evolutionary relationships between the members of this family according to the genetic structure changes were also elucidated.The expression patterns of CesA family members in response to drought stress were analyzed systematically by RNA sequencing and ribosome profiling,the duplication patterns of this family were analyzed from the perspective of replication,and the functions of CesA genes in maize were preliminarily explored based on the transcriptional regulatory network.【Result】There were 16 genes found in CesA family of maize,belonging to seven subfamilies according to the evolutionary relationship and three of them were newly discovered.The members had tissue specific and developmental period-specific responses to drought during development,and the effect of drought stress during seed development was larger than during plant growth.Members of the family interacted synergistically with each other in the synthesis of cellulose,with functional redundancy among them.【Conclusion】This study identified 16 maize CesA family members,and clarified the functional cooperation between each other and the response to drought stress during the seed development and plant growth.
