摘要 目的:观察蟾毒灵联合阿霉素对膀胱癌T24细胞生长的影响,探讨蟾毒灵联合阿霉素抗肿瘤作用的可能机制。方法:用不同浓度的蟾毒灵和阿霉素分别作用于T24细胞,用MTT法检测细胞增殖抑制率,流式细胞仪检测细胞凋亡率,RT-PCR检测凋亡基因Bax及Bcl-2表达水平,再联合使用两组药物作用T24细胞,检测相关凋亡指标。结果:MTT结果显示,蟾毒灵与阿霉素联合应用比单独应用阿霉素对T24细胞的增殖抑制率要明显提高,差异有统计学意义(P <0. 05);流式细胞仪结果显示,当蟾毒灵和阿霉素联合应用T24细胞的凋亡率要明显高于单独应用两种药物,差异有统计学意义(P <0. 05); RT-PCR结果显示,蟾毒灵联合阿霉素能明显下调凋亡抑制基因Bcl-2的表达,上调促凋亡基因Bax的表达,与单独用药相比差异有统计学意义(P <0. 05)。结论:蟾毒灵联合阿霉素可增敏阿霉素对T24细胞的增殖抑制及诱导细胞凋亡的能力;其抗肿瘤活性的机制可能与影响凋亡基因Bax及Bcl-2的表达有关。 Objective: To observe the effect of bufalin combined with adriamycin on the growth of bladder cancer T24 cells and explore the possible mechanism of bufalin combined with adriamycin antitumor effect. Methods: Different concentrations of bufalin and adriamycin were applied to T24 cells respectively. MTT assay was used to detect the rate of cell proliferation inhibition. Flow cytometry was used to detect the apoptosis rate. The expression of Bax and Bcl-2 T24 cells were treated with two groups of drugs,and the relative apoptosis indexes were detected. Results: The results of MTT showed that the combination of bufalin and adriamycin significantly increased the proliferation inhibition rate of T24 cells compared with adriamycin alone,the difference was statistically significant( P < 0. 05). Flow cytometry showed that when the combination of bufalin and adriamycin T24 cell apoptosis rate was significantly higher than the single application of two drugs,the difference was statistically significant( P < 0. 05). RT-PCR results showed that bufalin combination of adriamycin can significantly down-regulate the expression of apoptosis-inhibiting gene Bcl-2 and up-regulate the expression of pro-apoptotic gene Bax,which is significantly different from that of drug alone( P < 0. 05). Conclusion: Bufalin combined with adriamycin can enhance the sensitivity of adriamycin to the proliferation inhibition of T24 cells and induce apoptosis. The mechanism of anti-tumor activity may be related to the effects on the expression of Bax and Bcl-2.
