摘要
目的筛选中药罗仙子小分子多肽中的抗氧化活性部位并对其抗氧化活性进行初步评价。方法利用薄层-生物自显影技术筛选和分离罗仙子小分子多肽中抗氧化活性部位;测定该抗氧化活性部位对DPPH自由基、OH自由基和ABTS自由基的清除率;以H_2O_2氧化损伤人脐静脉内皮细胞ECV304为模型,测定罗仙子抗氧化活性部位对内皮细胞SOD、GSH-Px活力及MDA含量的影响。结果根据DPPHTLC法筛选到罗仙子小分子多肽抗氧化活性部位主要集中S1部位,该部位具有较好的清除自由基能力; S1部位可提高H_2O_2氧化损伤ECV304细胞的SOD、GSH-Px活力,降低MDA含量。结论 TLC-DPPH技术可有效筛选和分离罗仙子小分子多肽中抗氧化活性部位,且该部位具有较好的体外抗氧化活性。
Objective To screen and evaluate the antioxidant sites of low molecular weight peptides of Musca domestica maggot.Methods The antioxidant fractions of low molecule polypeptides from Musca domestica maggot were screened and separated by TLC-Bioautography,and the scavenging rate of DPPH,OH and ABTS free radicals was measured.Effect of the antioxidant fractions of Musca domestica maggot on endothelial cells SOD,GSH-PX and MDA was determined in ECV304which induced by H2O2.Results According to TLC-DPPH screening,the antioxidant active fraction of low molecule polypeptide from Musca domestica maggot was mainly located in S1,which had good free radical scavenging ability.S1could increase the SOD,GSH-PX activity and decrease the content of MDA in ECV304cells oxidized by H2O2.Conclusion TLC-DPPH technology can effectively screen and isolate the antioxidant active fraction of low molecule polypeptides of Musca domestica maggot,and this fraction has good antioxidant activity in vitro.
