摘要
目的通过比较子宫肌瘤组织与瘤旁正常子宫肌肉组织的细胞凋亡情况、凋亡相关蛋白的表达或活化情况,探讨子宫肌瘤的发生、发展与细胞凋亡的关系。方法收集20例子宫肌瘤瘤体及瘤体旁正常平滑肌组织、子宫肌瘤组织为研究组,同源正常子宫平滑肌组织为对照组。采用DNA片段化分析子宫肌瘤瘤体及瘤体旁正常平滑肌组织染色体DNA,采用Western Blot分析子宫肌瘤瘤体及瘤体旁正常平滑肌组织中B细胞淋巴瘤/白血病-2(Bcl-2)、Bcl-2相关X蛋白(Bax)的表达及Bcl-2/Bax比值,分析含半胱氨酸的天冬氨酸蛋白水解酶-3(Caspase-3)和Caspase-9的活化情况。结果研究组有5例出现DNA ladder条带,对照组有15例出现DNA ladder条带。研究组Bcl-2表达量为(0. 823±0. 102),显著高于对照组的(0. 348±0. 067);研究组Bax表达量为(0. 185±0. 073),显著低于对照组的(0. 429±0. 088);研究组Bcl-2/Bax比值为(4. 450±0. 760),显著高于对照组的(0. 810±0. 370)(P <0. 05)。与对照组相比,研究组中Caspase-3及Caspase-9的活化明显较少。结论子宫肌瘤的发生发展可能与细胞凋亡抑制有关,这一过程可能涉及凋亡相关蛋白Bcl-2及Bax的表达情况、Bcl-2/Bax比值变化以及Caspase-3及Caspase-9的活化。
Objective To investigate the relationship between the occurrence,development of uterine leiomyoma and apoptosis by comparing the apoptosis condition,the expression or activation of apoptosis-related proteins between uterine leiomyoma and normal uterine muscle tissues adjacent to the tumor.Methods Totally 20 cases of uterine leiomyoma and normal smooth muscle tissue adjacent to uterine leiomyoma were collected.The uterine leiomyoma tissue was designed as the research group,and the homologous normal uterine smooth muscle tissue was designed as the control group.DNA fragmentation was used to analyze the chromosomal DNA of uterine leiomyoma and normal smooth muscle tissues adjacent to the leiomyoma.Western Blot was used to analyze the expression of B-cell lymphoma/leukemia-2(Bcl-2),Bcl-2-related X protein(Bax)and the ratio of Bcl-2 to Bax in uterine leiomyoma and normal smooth muscle tissues adjacent to the leiomyoma.Activations of Caspase-3 and Caspase-9 were analyzed.Results DNA ladder bands were found in 5 cases in the research group and 15 cases in the control group.The expression of Bcl-2 in the research group was(0.823±0.102),which was significantly higher than(0.348±0.067)in the control group(P<0.05).The expression of Bax in the research group was(0.185±0.073),which was significantly lower than(0.429±0.088)in the control group(P<0.05).The ratio of Bcl-2 to Bax in the research group was(4.450±0.760),which was significantly higher than(0.810±0.370)in the control group(P<0.05).Compared with the control group,the activation of Caspase-3 and Caspase-9 in the research group was obviously less.Conclusion The occurrence and development of uterine leiomyoma may be related to the inhibition of apoptosis,which may involve the expression of Bcl-2 and Bax,the change of ratio of Bcl-2 to Bax and the activation of Caspase-3 and Caspase-9.
