摘要
目的探讨栀子苷对乳腺炎小鼠乳腺组织及乳腺上皮细胞中IL-6、TNF-α和IL-1β表达的影响。方法将40只分娩后雌性小鼠随机分为空白对照组、模型组、L组、M组和H组。建模后12 h,L组给予低剂量栀子苷治疗,M组给予中剂量栀子苷治疗,H组给予高剂量栀子苷治疗,空白对照组和模型组给予等量磷酸缓冲盐溶液。建模后24 h观察干预效果。结果模型组、L组、M组和H组TNF-α、IL-1β、IL-6 mRNA水平显著高于空白对照组(P<0.05);L组、M组和H组TNF-α、IL-1β、IL-6 mRNA水平显著低于模型组(P<0.05);M组和H组TNF-α、IL-1β、IL-6 mRNA水平显著低于L组(P<0.05);H组TNF-α、IL-1β、IL-6 mRNA水平显著低于M组和L组(P<0.05)。L组、M组和H组TNF-α、IL-1β、IL-6蛋白水平显著低于模型组(P<0.05);M组和H组TNF-α、IL-1β、IL-6蛋白水平显著低于L组(P<0.05);H组TNF-α、IL-1β、IL-6蛋白水平显著低于M组和L组(P<0.05)。结论栀子苷可抑制脂多糖诱导的乳腺炎症损伤,有降低TNF-α、IL-1β和IL-6炎症因子水平的作用。
Objective To study the effect of geniposide on expression of IL-6,TNF-α,and IL-1βin mammary glands and mammary epithelial cells of mastitic mice.Methods Forty female mice after childbirth were randomly divided into blank control,model,L,M,and H groups.At 12 h after modeling,the L group was treated with low-dose geniposide,the M group was administered intermediate-dose geniposide,the H group was treated with high-dose geniposide,and blank control and model groups were administered an equal amount of phosphate-buffered saline.At 24 h after modeling,the treatment effects were observed.Results mRNA expressions of TNF-α,IL-1β,and IL-6 in model,L,M,and H groups were significantly higher than that in the blank control group(P<0.05).Expressions of TNF-α,IL-1β,and IL-6 mRNAs in the L,M,and H groups were significantly lower than that in the model group(P<0.05).Expressions of TNF-α,IL-1β,and IL-6 mRNAs in M and H groups were significantly lower than that in the L group(P<0.05).mRNA expressions of TNF-α,IL-1β,and IL-6 in the M group were significantly lower than that in M and L groups(P<0.05).The levels of TNF-α,IL-1β,and IL-6 in L,M,and H groups were significantly lower than those in the model group(P<0.05).The levels of TNF-α,IL-1β,and IL-6 in M and H groups were significantly lower than those in the L group(P<0.05).The levels of TNF-α,IL-1β,and IL-6 in the H group were significantly lower than those in M and L groups(P<0.05).Conclusions Geniposide inhibits lipopolysaccharide-induced mammary gland inflammation and reduces the levels of proinflammatory factors TNF-α,IL-1β,and IL-6.
