摘要
目的探讨川芎提取物对^(60)Coγ电离辐射损伤小鼠的保护作用,及其对损伤小鼠脾脏组织细胞凋亡相关蛋白表达的影响。方法小鼠随机分为空白组(Normal)、辐照组(ionizing radiation,IR)、氨磷汀组(AMF,腹腔注射260 mg·kg^(-1))和川芎提取物组(LCXE1、LCXE2、LCXE3,依次灌胃给药50、100、200 mg·kg^(-1))。预防性给药3 d后,采用5 Gy ^(60)Coγ射线辐照建立小鼠电离辐射损伤模型。采用TUNEL标记和免疫组织化学染色法,观察川芎提取物对小鼠脾脏组织凋亡细胞数目及凋亡相关蛋白Bax、Bcl-2、p53表达的影响。结果与IR组相比,AMF明显降低脾组织凋亡细胞数量(P<0.01),川芎提取物剂量依赖性降低脾组织凋亡细胞数量(P<0.05);AMF、LCXE2、LCXE3能明显提高Bcl-2蛋白表达(P<0.01),明显降低Bax蛋白表达(P<0.05)及p53蛋白表达(P<0.01)。结论川芎提取物对^(60)Coγ射线损伤的小鼠有明显的改善作用,其作用机制可能为作用于细胞凋亡相关通路。
Aim To investigate the protective effect of Ligusticum chuanxiong Hort.extracts(LCXE)against cobalt 60-induced ionizing radiation injury in mice,and its effect on the expression of apoptosis-related proteins in splenic tissues of mice injured by ionizing radiation.Methods The model of ionizing radiation damage in mice was established by irradiation with 60 Coγrays.All mice were randomly divided into six groups,namely normal group,ionizing radiation(IR)group,amifostine(AMF)group and three LCXE groups(LCXE1,LCXE2 and LCXE3).Prophylactic administration had been continuously performed for three days before mice were given 60 Coγrays,of which normal and IR groups were given an equal volume of saline,AMF group were intraperitoneally injected of AMF at a dose of 260 mg·kg^-1,and the LCXE1,LCXE2,LCXE3 groups were intragastrically administered at the concentrations of 50,100,200 mg·kg^-1,respectively.After 30 minutes of the administration on the third day,the mice,except those in normal group,were all under the exposure of 60 Coγrays irradiation.The total irradiation dose was 5 Gy.According to the damage model,TUNEL assay and immunohistochemical staining were used to observe the effect of LCXE on the number of apoptotic cells and the expression of the apoptosis-related Bax,Bcl-2 and p53 proteins in splenic tissues.Results Compared with IR group,AMF decreased the number of apoptotic cells in splenic tissues(P<0.01),and LCXE decreased the number of apoptotic cells in splenic tissues,which was dose-dependent(P<0.05).Compared with IR group,AMF,LCXE2 and LCXE3 increased the expression of Bcl-2 protein(P<0.01),significantly decreased the expression of Bax protein(P<0.05)and the expression of p53(P<0.01).Conclusion It is speculated that LCXE could protect mice from 60 Coγradiation damage through the apoptosis pathway from ionizing radiation injury.
