摘要
目的探讨细胞周期依赖激酶抑制剂3(CDKN3)在胰腺癌中的表达及意义。方法采用免疫组化及Western blot法检测29例胰腺癌及癌旁组织CDKN3蛋白的表达,分析其与胰腺癌的临床病理参数关系;运用siRNA技术敲低胰腺癌细胞CDKN3表达,检测其对癌细胞迁移能力的影响。结果 CDKN3在胰腺癌组织中的表达水平显著高于癌旁组织,敲低CDKN3表达能显著降低胰腺癌细胞的迁移能力;胰腺癌患者CDKN3的表达水平与其淋巴结转移及TNM分期相关。结论 CDKN3是胰腺癌新的原癌基因,且高CDKN3表达提示胰腺癌患者预后不良。
Objective To investigate the expression of cyclin?dependent kinase inhibitor 3(CDKN3)in pancreatic cancer and its significance.Methods The specimens of pancreatic cancer and its adjacent tissues from 29 patients were collected.Expression of CDKN3 was detected by using immunohistochemistry and Western blot.The relationship between CDKN3 expression and clinical pathologic characteristics in pancreatic cancer was explored.Knockdown of CDKN3 in Panc?1 cell lines by transiently transfected with small interfering RNA drastically attenuated the migration of those cells.Results As compared with those in the adjacent tissues,CDKN3 was upregulated in the pancreatic cancer.The expression level of CDKN3 was correlated with lymphatic metastasis and TNM stage.Conclusions CDKN3 is a new oncogene of pancreatic cancer.Higher CDKN3 expression suggests poorer prognosis in patients with pancreatic cancer.
