H2S对NaCl胁迫下草木樨幼苗生理指标及抗氧化酶活性的影响 被引量:9

摘要 为了探究硫化氢(H_2S)增强牧草抗NaCl胁迫的能力及剂量效应,筛选适宜的喷施浓度。以草木樨(Melilotus suaveolens)为材料,采用水培的方法,用H_2S供体硫氢化钠(NaHS)处理NaCl胁迫下的草木樨幼苗,研分析H_2S对NaCl胁迫下草木樨幼苗叶片相对含水量、相对电导率、渗透调节物质、抗氧化酶活性、丙二醛(MDA)含量以及叶绿素含量的影响,研究H_2S对NaCl胁迫下草木樨幼苗的缓解效应机制。结果表明,适宜浓度NaHS处理提高了NaCl胁迫下草木樨幼苗叶片相对含水量,降低了叶片相对电导率;缓解了游离脯氨酸、可溶性糖含量的升高,促进可溶性蛋白含量增加;增强超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)活性,降低丙二醛含量,增加叶片中的叶绿素含量。不同浓度NaHS处理对缓解草木樨的NaCl盐胁迫效果差异显著(P<0.05),随浓度的增加,各指标均显示出一定的剂量效应,采用隶属函数法综合比较不同浓度NaHS处理后草木樨的生理及酶活性响应,0.1mmol·L-1的NaHS处理对NaCl胁迫的缓解效果最佳。 In order to examine the potential use and optimal dose of hydrogen sulfide(H 2S)to ameliorate NaCl stress in forage,the adequate treatment concentration was determined.Sweet clover(Melilotus suaveolens)was used in this study by applying a hydroponic method.Seedlings under NaCl stress were treated with NaHS to assess the effect of H 2S treatments on relative water content,relative conductivity,osmotic substance,antioxidant enzyme activity,and malondialdehyde(MDA)content of sweet clover seedlings leaves under NaCl stress to examine the relief mechanisms in sweet clover seedlings under NaCl stress treated with H 2S.The results showed that the relative water content of sweet clover seedling leaves under NaCl stress increased following NaHS treatment,whereas relative conductivity decreased;the increase of free proline and soluble sugar content was mitigated,and the content of soluble protein increased;the activities of superoxide dismutase(SOD),peroxidase(POD),and catalase(CAT)increased,the MDA content decreased,and leaf chlorophyll content increased following the treatment.The observed changes in the tested indexes showed a certain dose effect.A comprehensive comparison of different physiological responses and enzymatic activities in sweet clover after NaHS treatment was performed using a membership function,and indicated that 0.1 mmol·L-1 NaHS treatment produced the best effect in NaCl-stressed plants.
