摘要
【目的】探讨‘苦水玫瑰’花色苷最佳提纯工艺参数及对几种常见致病菌的抑菌效果.【方法】采用有机溶剂浸提法,在单因素试验基础上,运用Box-Behnken响应面分析优化‘苦水玫瑰’花色苷最佳提取工艺条件,经D-101型大孔树脂的动态纯化工艺后,确定最佳纯化工艺参数,测定其纯度,并进一步运用滤纸片法探究其抑菌效果.【结果】‘苦水玫瑰’花色苷最优提取工艺条件:提取液pH=1.05、乙醇浓度81.5%、提取时间45min、料液比1∶90(g/mL),提取2次,提取率4.27 mg/g.最佳动态吸附条件:上样液pH=2,吸附流速1 mL/min,上样液浓度0.15mg/mL,吸附率76.25%,95%酸性乙醇将花色苷全部洗脱.纯化后的花色苷对大肠杆菌和沙门氏菌抑菌效果不明显,对金黄色葡萄球菌抑菌明显,其抑菌圈直径为(6.35~10.12)±0.05 mm,最低抑菌浓度为1 mg/mL.【结论】利用响应面法确定最佳提取工艺条件合理,可用于‘苦水玫瑰’花色苷的提取.苦水玫瑰花色苷纯度由原来的5.862%提高到了23.97%,纯化效果显著,对金黄色葡萄球菌有明显抑菌效果.
【Objective】To explore the optimal extraction and purification process parameters of anthocyanins from Rosa rugosa and its antibacterial activity against several common pathogenic bacteria.【Method】Using organic solvent extraction method,based on the single factor experiment,the optimal extraction conditions of anthocyanins from Rosa rugosa were optimized by Box-Behnken response surface analysis,the optimum purification conditions were determined by the dynamic purification of D-101 macroporous resin Process parameters,determination of its purity,and further use of filter paper to explore its antibacterial effect.【Result】The optimal extraction conditions of anthocyanins from Rosa rugosa were pH=1.05,ethanol concentration 81.5%,extraction time 45min,ratio of solid to liquid 1∶90(g/mL),extract twice,extraction rate 4.27 mg/g.The optimal dynamic adsorption conditions were as follows:the sample solution pH=2,the adsorption flow rate 1 mL/min,the sample concentration 0.15mg/mL,the adsorption rate 76.25%,and all of the anthocyanins eluted with 95%.The purified anthocyanins had no obvious antibacterial effect on Escherichia coli and Salmonella,and showed obvious antibacterial activity against Staphylococcus aureus.The diameter of antibacterial circle was(6.35~10.12)±0.05 mm and the minimum inhibitory concentration was 0.5 mg/m L.【Conclusion】Response surface method to determine the best extraction process conditions can be used for the extraction of anthocyanins from Rosa rugosa.Anthocyanins from Rosa rugosa purity from the original 5.862%to 23.97%,the purification effect was significant,Staphylococcus aureus obvious inhibitory effect.
