Construction of Nanoparticle Drug Delivery System Based on Mesoporous Silica

摘要 利用阳离子表面活性剂十六烷基三甲基溴化铵(CTAB)为模板剂,正硅酸乙酯(TEOS)为硅源,采用水热合成法、酸液萃取法制备介孔二氧化硅纳米颗粒(MSNs)。氮气吸附—脱附实验结果表明,所制备的MSNs为典型的介孔结构,孔径分布均匀,适合于药物载带。抗肿瘤药物姜黄素(Cur)的载带与释放表明MSNs为典型的pH响应药物载体,在pH=7.4时Cur的包封率和载药率分别高达50%和15%左右,而在pH=5.0的环境中则快速释放药物。 The mesoporous silica nanoparticles(MSNs)are prepared by adopting cetyl trimethyl ammonium bromide(CTAB)template and tetraethyl orthosilicate(TEOS)as silicon source.The N2 adsorption/desorption isotherms demonstrated the MSNs exhibit typical IV-typed isotherms with H1-hysteresis loops,which are the properties of typical mesoporous materials.The anti-cancer drug curcumin(Cur)storage/release properties were demonstrated to be pH-responsive,in which,the Cur encapsulation efficiency(EE,%)and drug loading content(DLC,%)could up to over 50%and 15%at pH7.4,respectively.However,the drug release might be beneficial at the reduced pH of 5.0.
