摘要
为得到高质量提取麦曲总DNA的方法,采用SDS法、氯化苄法、CTAB法、超声波法、Soil DNA kit法、SDS高盐法及SDS-CTAB法对黄酒麦曲中总DNA的提取效果进行了比较,通过凝胶电泳、PCR、紫外分光光度计及Real-time PCR对不同方法提取产物进行分析得出7种方法中SDS-CTAB法对于提取麦曲总DNA效果最好,蛋白、多糖及小分子等污染较少,DNA提取的质量浓度达到149.6 ng/μL,相对于SDS法,细菌和真菌模板数分别达到其2.343倍和1.753倍。
In order to isolate high quality DNA from Wheat Qu the effect of DNA isolation by seven different methods were evaluated.The extracted DNA was analysised by gel electrophoresis,PCR,ultraviolet spectrophotometry,and real-time fluorescent PCR,respectively.The results showed that SDS-CTAB method has the best effect on the isolation of total DNA of Wheat Qu.This method'products showed less contamination from protein and polysaccharide,the yield of the DNA was reached 149.6 ng/μL and its'relative account of template is almost 2.343 and 1.753 times in bacteria and fungi respectively compared to SDS method.
