摘要
拟南芥是基因功能研究的模式植物,其DNA提取是突变体筛选与鉴定的必要环节。本研究利用FTA卡便能快捷地提取拟南芥叶片DNA。通过与CTAB法和试剂盒法提取DNA相比较:FTA卡法一方面操作简单、省时,成本低;另一方面其提取的DNA为模板,PCR扩增得到的条带同样清晰、完整、得率高;此外,FTA卡上的DNA可在室温下长久保存,且比EP管更节省存储空间。一个完整的叶片即可以进行上百次PCR反应,适合应用到基因图位克隆、突变体批量筛选与鉴定等实验,这将极大地节约实验成本、劳力以及时间,加快实验进程。
Arabidopsis thaliana is widely used as a model system in plant gene function studies. DNA extraction is an essential step in mutant screening and identification. By FTA card method, DNA templates of Arabidopsis thaliana could be extracted quickly. Compared with CTAB method and Reagent Kit method to extract DNA: FTA card method had the advantages of simple operation, time saving and low cost; on the other hand, using DNA samples extracted with FTA card as template, the band obtained by PCR amplification was clear, complete and high yield; furthermore, on the FTA card, DNA could be stored for a long period at room temperature, and its storage need less space than EP tube. A complete leaf is enough for PCR reaction hundreds of times, is suitable to be applied in experiments like genetic map spaces cloning, mutant batch screening and identification, which might greatly save the experimental cost, labor, time, and shorten the experimental process as well.
