摘要
目的探讨加味脑泰方对卵巢摘除脑缺血大鼠神经细胞凋亡的影响及其机制。方法雌性Sprague-Dawley大鼠40只随机分为假手术组(n=10)、模型组(n=10)、雌激素组(n=10)和加味脑泰方组(n=10)。模型组、雌激素组、加味脑泰方组大鼠摘除卵巢。去卵巢术后11 d,雌激素组、加味脑泰方组分别予雌激素和加味脑泰方灌胃3 d;术后14 d,模型组、雌激素组、加味脑泰方组线栓法制备局灶性脑缺血模型。缺血24 h后取脑组织,TUNEL法观察神经细胞凋亡率,Western blotting检测海马细胞外信号调节蛋白激酶1/2(ERK1/2)、c-Jun氨基末端蛋白激酶(JNK)蛋白活化程度。结果与模型组比较,加味脑泰方组和雌激素组神经细胞凋亡率显著降低(P<0.001),ERK1/2蛋白活化程度明显升高(P<0.01),p-JNK蛋白活化程度明显降低(P<0.01)。结论加味脑泰方可能通过提高p-ERK1/2蛋白活化,降低p-JNK蛋白活化,降低脑缺血后神经细胞凋亡率。
Objective To investigate the effect of Jiawei Naotaifang on neuronal apoptosis and the mechanism in ovariectomized rats with cerebral ischemia.Methods Female Sprague-Dawley rats(n=40)were randomly divided into sham group(n=10),model group(n=10),estrogen group(n=10)and Jiawei Naotaifang group(n=10).The model group,estrogen group and Jiawei Naotaifang group were ovariectomized.Eleven days after ovariectomy,the estrogen group and Jiawei Naotaifang group were given estrogen and Jiawei Naotaifang respectively intragastrically for three days.14days after ovariectomy,the model group,estrogen group and Jiawei Naotaifang group were modeled cerebral ischemia with Langa's method.24hours after modeling,the apoptosis rate of neurons was detected with TUNEL,and the activation of extracellular signal-regulated kinase1/2(ERK1/2)and c-Jun N-terminal kinase(p-JNK)in hippocampus were detected withWestern blotting.Results Compared with the model group,the apoptosis rates decreased in Jiawei Naotaifang group and the estrogen group(P<0.001),with more activation of ERK1/2(P<0.01)and less activation of JNK(P<0.01).Conclusion Jiawei Naotaifang can protect neuron from apoptosis by promoting the activation of ERK1/2and inhibiting the activation of p-JNK.
