摘要
目的观察左归丸对去卵巢小鼠的雌激素样作用。方法采用经典的双侧卵巢切除法使小鼠体内雌激素降低,术后3天,左归丸灌胃给药4周,取材,测血清中雌二醇水平、计算子宫系数。通过HE染色,观察阴道和子宫形态改变,并对比子宫和阴道内膜的厚度的前后变化。结果子宫系数:左归丸高剂量组与模型组比较,具有显著性差异(P<0.05);子宫内膜:左归丸高、中剂量组与模型组比较均具有极显著性差异(P<0.01);阴道内膜:左归丸高、中、低计量组与模型组比较均具有极显著性差异(P<0.01);可以明显缓解子宫和阴道内膜的萎缩;血清中雌激素水平:左归丸高剂量组与模型组相比,小鼠血清中的雌二醇含量并没有明显性变化(P>0.05),左归丸中剂量、低剂量组与模型组相比,小鼠血清中的雌二醇水平回升趋势具有显著性差异(P<0.05)。结论左归丸具有明显的类雌激素样作用,能够改善雌激素减少导致的子宫萎缩和阴道形态的变化,但左归丸提高去卵巢小鼠血清中的雌激素含量与其类雌激素样作用并没有明显相关性。
Objective To observe the estrogen-like effects of Zuogui pill on ovariectomized mice.Methods The classic bilateral ovariectomy was conducted in mice to reduce the estrogen in mice.Three days after surgery,Zuogui pills gavaged for4weeks.Then serum estradiol levels was measured and the uterus coefficient was calculated.The morphological changes of the vagina and uterus were observed by HE staining,and the changes of the thickness of the uterus and vaginal intima were compared.Results Uterine coefficient:There was significant difference between the Zuogui pills high dose group and the model group(P<0.05).Uterus endometrium:There was significant difference between the model group and the Zuogui pills high and middle dose group(P<0.01).Vaginal endometrium:There was significant difference the Zuogui pills high,middle,and low dose group compared with the model group(P<0.01).Estrogen levels in the serum:compared with the model group,there was no significant change in Zuogui pills high dose group(P>0.05).While,there was significant change in middle and low dose group compared with the model group(P<0.05).Conclusion Zuogui pill has an obvious estrogen-like effect,which can improve the atrophy of uterus and the change of vagina form.However,this experiment shows that Zuogui pill has no significant correlation with estrogen-like effect in serum of ovariectomized mice,so its mechanism remains to be further studied.
