黄果槲寄生果实多糖对环磷酰胺诱导免疫低下小鼠免疫功能影响 被引量:13

摘要 【目的】用腹腔注射环磷酰胺(CTX)成功建立免疫低下小鼠动物模型,以黄果槲寄生果实多糖(Viscum coloratum(Kom.)Nakai f.lutescens kitag polysaccharide,VCP)为材料,研究多糖对免疫低下小鼠免疫功能的影响.【方法】将建模成功的小鼠随机分为免疫低下模型组、VCP高、中、低剂量处理组,以生理盐水为对照组.连续灌胃给药4周,研究不同剂量的VCP对免疫低下小鼠的胸腺指数、脾脏指数和炭廓清指数、小鼠免疫脏器指数和脾脏淋巴细胞活性、以及肝组织GSH-Px活力和血清中IL-2、IL-4、IFN-γ生成水平、MDA含量及SOD活力等指标的影响.【结果】VCP高剂量能显著提高模型小鼠脾脏指数、脾淋巴细胞增殖作用、廓清指数、吞噬指数、半数溶血值及足趾厚度差;VCP低、中、高剂量促使IL-2、IL-4、IFN-γ生成水平、GSH-Px活力和SOD活力明显提高,MDA含量明显降低.【结论】黄果槲寄生果实多糖对CTX致免疫低下小鼠的免疫功能具有增强作用,同时可以提高机体抗氧化能力和延缓衰老的作用. 【Objective】The study was to investigate the effects of V.coloratum polysaccharide(VCP)on the immunity activity of immunodepressive mice induced by cyclophosphamide(CTX).【Method】The Kunming mice were randomly divided into five groups,including immunodepressive model group,and VCP-treated groups(low-,medium-,and high-dosage),with normal saline as the control group.The VCP was performed intragastric administration of polysaccharide for four weeks,to investigate the effects of polysaccharides on indexes of thymus and spleen,spleen lymphocyte proliferation,GSH-Px activity,clearance index,phagocytic index,HC50,the level of serum IL-2,IL-4,IFN-γ,MDA contents and SOD activity and toe thickness difference in CTX-induced immunodepressive rats.【Result】Compared with CTX group,indexes of spleen,clearance,phagocytic,spleen lymphocyte proliferation,HC50and toe thickness difference in VCP groups with high dosage were promoted significantly.All VCP groups could significantly increased the level of serum IL-2,IL-4,IFN-γand GSH-Px activity,SOD activity CTX-induced immunodepressive mice and decreased the content of MDA in serum.【Conclusion】The results suggested that VCP could enhance immunologic functions of immunodepressive mice induced by CTX,improve the body's antioxidant capacity scavenging free radicals and take anti-aging effect.
