摘要
[目的]研究葛花醒酒护肝方水提取物对醉酒小鼠的解酒护肝作用及其机制,并评价其急性毒性。[方法]建立小鼠醉酒及酒精中毒模型,给予葛花醒酒护肝方水提取物高、中、低剂量干预,观察并记录动物醉酒(翻正反射消失)潜伏期、醉酒时白酒摄入量、死亡时间和死亡时白酒摄入量;同时测定动物血清中ALT、AST活性以及肝脏组织中GSH、MDA、ADH的活性;以成人拟用剂量的1 000倍连续给予动物灌胃给药14 d,观察该期间内发生的不良反应,测定受试动物对该制剂的最大耐受量。[结果](1)葛花醒酒护肝方水提取物高、中剂量组可显著延长小鼠的醉酒潜伏期、死亡时间和增加小鼠醉酒时的白酒摄入量、死亡时白酒的摄入量(P<0.05或(P<0.01),表现出明显的解酒作用及一定的量效关系。(2)葛花醒酒护肝方水提取物的高、中剂量组显著降低小鼠血清中ALT、AST活性,显著降低肝组织中MDA活性,明显提高肝组织中GSH和ADH的活性(P<0.05或(P<0.01)。(3)连续给药期间,受试动物的体形、体重、体重增长、摄食量及摄食量变化率未见异常,小鼠对葛花醒酒护肝方水提取物的经口最大耐受量大于48 g/(kg·d)。[结论]葛花醒酒护肝方水提取物具有明显的解酒护肝作用,其解酒护肝作用与提高肝脏中GSH、ADH的活性以及降低血清中ALT、AST的活性有关;葛花醒酒护肝方水提取物用于解酒安全而稳定。
[Objective]To study the anti-alcohol and liver-protective function and mechanism of water extract from Gehua Xingjiu formula in over-drunk mice,and evaluate the acute toxicity of the formula.[Methods]The mice model of over-drunk and alcoholism was established to be intervened with high,middle,low doses of water extract from Gehua Xingjiu formula.The incubation period of animal drunk(disappearance of right reflex),alcohol intake during drunkenness,time of death and liquor intake at death were observed and recorded.Meanwhile,the activity of ALT and AST in animal serum and the activity of GSH,MDA and ADH in liver tissue were determined.The mice were given gavage administration of1000times of the intended dose for adults for14days continuously.The adverse reactions during the period were observed,and the maximum tolerated dose of the preparation for the test animals was determined.[Results]①The high,middle dose groups of water extract from Gehua Xingjiu formula could significantly prolong the latent period of drunkenness,the time of death of mice and increase the intake of alcohol when mice drunk,the intake of alcohol when mice died(P<0.05or P<0.01),showing obvious anti-alcohol effect and a certain dose-effect relationship.②The high,middle dose groups of water extract from Gehua Xingjiu formula could significantly decrease the activity of ALT and AST in serum,reduce the activity of MDA in liver tissue and could obviously increase the activity of GSH and ADH in liver tissue in mice(P<0.05or P<0.01).③During continuous administration,the body shape,weight,weight gain,food intake and rate of change of food intake of the test animals showed no abnormalities.The maximum tolerated oral dose of water extract from Gehua Xingjiu formula for mice was greater than48g/(kg·d).[Conclusion]The water extract from Gehua Xingjiu formula has obvious anti-alcohol and liver-protective function,which is related to increase of the activity of GSH and ADH in liver and decrease of the activity of ALT and AST in serum.The extract of the
