3个群体方斑东风螺线粒体COI基因的遗传多样性分析 被引量:4

摘要 采用PCR产物直接测序法对来自福建东山、广东湛江和广西北海的3个群体方斑东风螺(Babylonia areolata)的COI基因进行比较分析。结果表明:COI基因片段长度为680 bp,碱基组成显示A+T比例较高,约为62.42%;种群间多态位点数为32,单倍型多样性指数为0.707±0.092,核苷酸多态性指数为0.001 85±0.000 39,平均核苷酸差异数为1.252。以泥东风螺(Babylonia lutosa)和织纹螺(Nassarius)为外群,构建了NJ系统发育树,其拓扑结构显示,3个群体的方斑东风螺首先聚类到一起,然后与泥东风螺聚为一支,与织纹螺形成不同的分支。 We analyzed and compared the polymorphism of mitochondrial COI gene sequence derived from threedifferent groups of Babylonia areolata in Dongshan, Zhanjiang and Beihai by means of PCR. The results showed that thenucleotide sequence of COI gene was 680 bp with a high percentage of A+T base composition (about 62.42%); therewere 32 singleton poylmorphic sites, 0.707±0.092 haplotype diversity, 0.001 85±0.000 39 nucleotide polymorphismsindex, and 1.252 average number of nucleotide differences. Taking Babylonia lutosa and Nassarius as outgroups, we builtNJ phylogenetic tree, its topology suggested that all the Babylonia areolata clustered together first, then mixed withBabylonia lutosa for one, and finally Nassarius formed independent.
