几种因素对人外周血IL2产生的影响

摘要 作者采用人外周血淋巴细胞(PBL)分析了从PBL中除去巨噬细胞、较小剂量的γ射线(10Gy),消炎痛及Raji细胞(B淋巴母细胞白血病)等因素对PBL产生IL2的影响。结果表明,从PBL中除去巨噬细胞能使IL2产生明显下降,甚至无法测出IL2的活性。回加自体巨噬细胞可以恢复IL2的产生,当回加的巨噬细胞浓度超过15%时,对IL2的产生有抑制作用,但经10Gyγ射线照射的巨噬细胞对IL2产生的抑制作用减弱。前列腺素(PGE)的合成抑制剂消炎痛、Raji细胞及小剂量的γ射线能显著提高IL2的产生,但消炎痛和Raji细胞对经小剂量γ射线照射的PBL产生IL2的促进作用不明显。这些结果提示,巨噬细胞具有促进IL2产生和抑制IL2产生的双重作用。 In the report we studied the effects of Macrophages,lower dose of radation ,indomethacin,and Raji cells on IL2 production by human perpheral blook T lymphocytes. The results showed macrophages were strictly required for IL2 production at appropriate concentrations of PHA. The purified T lymphocytes produced undetectable IL2 after stimulation with PHA, but IL2 production by purified T lymphoxytes seems to be relativly indenpendent of macrophage at high concentrations of PHA (500-750μg/ml). When macroahages were added back to the culture,IL2 production was markedly increased with increasing percentages of macrophage. More than 15% of macrophage inhibited IL2 production but the inhibition led by irradiated macrophages (30Gy) was more weaker than that of no-irradiated macrophages. Indomethacin (inhibitor of prostaglandin synthesis), Raji cells and lower dose of Υ-ray enhanced IL2 production by human peripheral blood lymphocytes. However, macrophages indomethacin and Raji cells did not much affect IL2 production by irradiated human peripheral blool lymphocytes. There data suggest macrophages can both enhance and inhibit IL2 production by lymphocytes.
