摘要
目的对Yam1B细胞自发产生的一种可抑制人T、B细胞增殖的可溶性因子(Yam-SF)进行研究。方法3H-TdR掺入法进行淋巴细胞增殖抑制测定,ELISA法测定免疫球蛋白,间接免疫荧光法测定IL-2R,CTLL-2细胞3H-TdR掺入法测定IL-2活性。结果Yam-1B培养上清(Yam-SF)经pronase水解消化而失活,经56℃处理10分钟,或在pH10环境中处理6小时抑制活性丧失,在pH2.0环境中可保持其抑制活性。凝胶过滤试验表明,培养上清抑制活性分布在分子量43000~67000范围内,且TGF-β抗体不能阻断其抑制活性。结论Yam-SF可能是一种新的免疫调节因子,该因子可抑制人T细胞及B细胞的增殖,并抑制正常B细胞免疫球蛋白的产生,但对PHA剌激的人淋巴细胞IL-2的产生以及IL-2受体(IL-2R)的表达无抑制作用。
Yam 1B,a human B lymphoblastoid cell line, spontaneously produced an immunoregulatory factor,which suppressed the proliferation of human T and B lymphocytes and inhibited immunoglobulin generation by normal B cells.The serumfree Yam 1B culture supernatant inhibited neither the expression of IL2 receptor nor the production of IL2 in the lymphocytes stimulated with PHA. The inhibitory activity of Yam 1B supernatant(YamSF) was inactivated at 56℃ for 10 min and at pH10.0 for 6h and abrogated by digestion with pronase, but remained at pH2.0. Gelfiltration column assay showed a peak of inhibitory activity in the fraction containing molecules of apparent MW of 43kD to 67kD. The inhibitory activity was not blocked by the antiTGFbeta antibody. These results indicate that YamSF appear to produce a novel immunoregulatory factor.
