摘要
氨基酸分析方法是测定含游离氨基酸,多肽或蛋白质样品中氨基酸含量的方法。在过去几十年中,氨基酸分析主要是由经典的柱后衍生-离子交换色谱和柱前衍生-高效液相色谱完成的。随着现代氨基酸分析技术的发展,高通量和高灵敏性的质谱被广泛应用于氨基酸分析中,并发挥了重要的作用。作者就近年来国内外气相色谱、液相色谱和毛细管电泳与质谱联用技术在氨基酸分析中的应用进行了综述并比较了这几种氨基酸分析技术。
Amino acid analysis is used to determine the amino acid content of amino acid-,peptide-and protein-containing samples.In the last decades,amino acid separations have been carried out mainly by ion-exchange chromatography with post-column derivatization and RP-HPLC with pre-column derivatization.Along with the modern technique of amino acid analysis,high throughput and sensitivity mass spectrometry has been widely used in the analysis of amino acids and brings significant benefits.This review focus on the application of the gas chromatography,liquid chromatography and capillary electrophoresis in combination with mass spectrometry in the analysis of amino acids,for which comparison are made.
