摘要 建立了高效液相色谱法直接测定生物催化产物中β-丙氨酸含量的新方法。转化产物经过衍生试剂2,4-二硝基氟苯(DNFB)柱前衍生后进行液相色谱分析,色谱柱为Zorbax SB C18柱(5μm,4.6mm×250mm);流动相分别为0.02mol/LNaAc-HAc缓冲液(A,pH6.2)和流乙腈(B);线性梯度洗脱色谱柱,流动相B浓度(体积百分含量)0→25min内由5%升高至40%;洗脱液流速为1.0mL/min;检测波长为360nm,参比波长为600nm;样品进样量为20μL。 A new method for determination of β-alanine in biocatalysis process was established by combining Reversed Phase High Performance Liquid Chromatography (HLPC) with Preeolumn Derivatization. The samples were derivatized with 2,4-dinitro-1-fluorobenzene (DNFB) and analyzed by HPLC with a 5μm Zorbax SB C18 column at 30℃ using gradient elution with diode array detection at 360nm. The mobile phase A was NaAc-HAe buffer (pH6.2), and the mobile phase B was acetonitrile. From 0 to 25 minutes, the concentration (V/V) of mobile phase B increased from 5% to 40%. The flow rate was 1.0 mL · min^-1 and the injection volume was 20μL.
