摘要
植物组织培养中玻璃化现象的发生极为普遍,是影响植物离体快繁的重大障碍。本试验以柿树为材料,对正常苗与玻璃苗的形态学差异进行了观察,研究了玻璃化现象发生的原因及防治方法。结果表明:提高培养基中琼脂粉的含量(达10g/L),可降低培养基的水势,尤其是降低了试管苗的水势,能有效地防止玻璃化现象的发生,同时有利于继代苗的增殖和生长;适当减少每瓶中接种数目及缩短继代培养时间,也可降低玻璃苗的发生频率。
The frequent occurrence of the vitrified shoot was the important hinder in fast reproduction of persimmon (Diospyros kaki Linn. ) ,The morphology difference between normal and vitrified shoots by using persimmon as material was surveyed and the reason and methods to solve the problem were studied .The results showed that by increasing the content of agar (to 1%)in the medium could reduce the water portential in the medium, especiallg the water portential of tuber shoots. This could prevent occurrence from vitrified shoots and favor the reproduction and growth of subculture shoots,Also reducing the number of shoots in a bottle and shortening subculture period could decrease the vitrified shoot proportion.
