摘要
目的探讨TCEAL7、Wnt和β-catenin在结直肠癌组织中的表达情况及TCEAL7作为结直肠癌标志物的价值。方法选取50例结直肠癌术后患者,收集手术切除的结直肠癌组织和癌旁正常组织。采用逆转录聚合酶链反应(RT-PCR)检测TCEAL7基因的表达,采用免疫组织化学技术检测Wnt、β-catenin蛋白的表达。分析TCEAL7、Wnt、β-catenin表达情况的相关性及三者表达情况与结直肠癌患者临床特征的关系,TCEAL7与预后的关系。结果 TCEAL7在结直肠癌组织中的相对表达量明显低于癌旁正常组织(P<0.01)。不同临床分期、淋巴结转移情况、远处转移情况的结直肠癌患者的TCEAL7、Wnt、β-catenin的表达情况比较,差异均有统计学意义(P<0.05),但不同性别、年龄结直肠癌患者的TCEAL7、Wnt、β-catenin的表达情况比较,差异均无统计学意义(P>0.05)。结直肠癌组织中TCEAL7的表达情况与Wnt、β-catenin的表达情况均呈负相关(P<0.01)。TCEAL7高表达患者的生存情况优于TCEAL7低表达患者(P<0.05)。结论 TCEAL7、Wnt、β-catenin的表达情况可能与结直肠癌患者临床分期、淋巴结转移情况、远处转移情况有关,TCEAL7表达与Wnt、β-catenin表达均呈负相关,TCEAL7可以用于评估结直肠癌患者的预后情况,可作为结直肠癌标志物。
Objective To investigate the expression of TCEAL7, Wnt, and β-catenin in colorectal cancer tissues and the value of TCEAL7 as a marker of colorectal cancer. Method The colorectal cancer tissues and adjacent normal tissues were collected from 50 patients with primary colorectal cancer who had surgical treatment. The TCEAL7 gene expression was examined by reverse transcription polymerase chain reaction （RT-PCR）, and the expression of Wnt and β- catenin proteins were determined immunohistochemically. The correlation among the expression of TCEAL7, Wnt and β- catenin, and the relationship between these markers and the clinical characteristics, as well as the association between TCEAL7 and prognosis were analyzed. Result The relative expression of TCEAL7 in colorectal cancer tissues was lower than that in the adjacent normal tissues （P〈0.01）. The expression of TCEAL7, Wnt and β-catenin in colorectal cancer patients with different clinical staging, lymph node metastasis, and distant metastasis were significantly different （P〈 0.05）, but no differences with age and gender （P〉0.05）. The expression of TCEAL7 was negatively correlated with the expression of Wnt and β-catenin （P〈0.01）. Higher expression of TCEAL7 was associated with better survival compared with those with lower TCEAL7 expression （P〈0.05）. Conclusion The expression of TCEAL7, Wnt and β-catenin are related to clinical staging, lymph node metastasis, and distant metastasis, TCEAL7 expression is negatively associated with Wnt and β-catenin expression, and can be used to predict the prognosis in colorectal cancer patients. TCEAL7 can be as a marker of colorectal cancer.
