摘要
目的分析牙龈卟啉单胞菌阳性率及其rag基因型在慢性牙周炎合并糖尿病患者唾液中的分布情况,探讨糖尿病参与并影响牙周炎发病的可能机制。方法收集慢性牙周炎患者及合并糖尿病的慢性牙周炎患者各20例。采集受试者的唾液样本,PCR法定性检测唾液中牙龈卟啉单胞菌的检出率及其rag基因型,统计学分析两组受试者牙龈卟啉单胞菌及其rag基因型的检出差异。结果经16S rDNA PcR法检测,伴糖尿病的牙周炎患者唾液中牙龈卟啉单胞菌阳性检出率为[65%(13/20)]与慢性牙周炎组[70%(14/20)]相比无统计学差异(P>0.05),但rag-1基因型的检出率后者[45%(9/20)]显著高于前者[5%(1/20)],rag3在两组中均未检出,rag2和rag4在两组的检出未见差异。结论慢性牙周炎患者比合并糖尿病的慢性牙周炎患者唾液中更易检出rag-1型的牙龈卟啉单胞菌,提示可能与糖尿病患者唾液流量及成分改变对菌群的影响所致。
Objective Analysis of porphyromonas gingivalis positive rate and its rag genotype in saliva of patients with chronic periodontitis and diabetes mellitus for a better understanding of the relationship between diabetes and periodontitis.Methods Saliva samples were collected from 20 chronic periodontitis patients,20 chronic periodontitis with Type 2 Diabetes Mellitus .The occurrence of P.gingivalis was determined by polymerase chain reaction(PCR) using 16S rDNA-specific Primers . Analysis the rag genotypes of P.gingivalis positive sample by PCR.Results The positive rate of Porphyromonas gingivalis in saliva with diabetic periodontitis was 65% (13/20). There was no significant difference between the positive rate of Porphyromonas gingivalis in saliva with diabetic periodontitis and that in chronic periodontitis group (70% (14/20)) (P 〉 0.05). The positive rate of Rag-1 in periodontitis was 45% (9/20).The positive rate of Rag -1 in chronic periodontitis was signifcantly higher than that of the former.Rag-3 was not detected in the two groups .There was no difference between Rag-2 and rag-4 in the two groups.Conclusion Rag-1 was more easily detected in saliva of patients with chronic periodontitis than those of patients with chronic periodontitis and diabetes mellitus, suggesting that the changes of salivary fow and composition may be related to the infuence of bacterial fora in diabetic patients.
