摘要
目的探讨血清食欲素A(orexin-A)水平与阿尔茨海默病(AD)及睡眠质量的相关性.方法收集60例AD患者(AD组)和60例健康者(对照组),采用简易精神状态评价量表(MMSE)和匹兹堡睡眠质量指数量表(PSQI)分别评估两组研究对象的认知功能和睡眠质量;采用酶联免疫吸附测定法(ELISA)检测其血清orexin-A水平;对血清orexin-A水平与MMSE及PSQI量表评分之间进行相关性分析.结果AD组患者的血清orexin-A水平显著高于对照组(P〈0.05);AD组MMSE量表评分及各分项评分均显著低于对照组(P〈0.05);AD组的PSQI量表评分、入睡困难以及睡眠障碍评分均显著高于对照组(P〈0.05),而睡眠质量、睡眠时间、睡眠效率、催眠药物使用以及日间功能障碍评分与对照组比较,差异无统计学意义(P〉0.05).PSQI量表评分与MMSE量表评分呈负相关;而血清orexin-A水平与MMSE量表评分及各分项评分均呈负相关(r〈0,P〈0.05),与PSQI量表评分及各分项评分均呈正相关(r〉0,P〈0.05).结论血清orexin-A可能在AD的发生、发展中有一定作用,其机制可能与其对睡眠质量的影响有关.
Objective To explore the correlation between serum orexin-A level and Alzheimer's disease (AD) and sleep quality. Methods The cognitive function and sleep quality of 60 patients with Alzheimer's disease (AD group) and 60 healthy people (control group) were evaluated by mini-mental state examination (MMSE) and Pittsburgh sleep quality index (PSQI) respectively; the serum orexin-A level was detected by enzyme-linked immunosorbent assay (ELISA). The correlation between serum orexin-A level and MMSE and PSQI were analyzed. Results The serum orexin-A level of AD group was signifcantly higher than that of control group (P〈0.05); the MMSE score and each sub-item score of AD group were signifcantly lower than those of control group (P〈0.05); the PSQI score, falling sleep diffculty score, and sleep disorder score of AD group were all signifcantly higher than those of control group (P〈0.05), while there were no statistically signifcant difference in the scores of sleep quality, sleep time, sleep effciency, hypnotic drug use, and daytime dysfunction between AD group and control group (P〉0.05). PSQI score was negatively correlated with MMSE score, the serum orexin-A level was negatively correlated with MMSE score and each sub-item score (r〈0, P〈0.05), but positively correlated with PSQI score and each sub-item score (r〉0, P〈0.05). Conclusion Serum orexin-A may play a role in the occurrence and development of Alzheimer's disease, and its mechanism may be related to its effect on sleep quality.
