水凝胶对大鼠Ⅱ度放射性皮炎创面愈合影响机制探讨 被引量:3

Mechanism of Urgo Hydrogel on Ⅱ degree radiation-induced dermatitis in rats

摘要 目的放射性皮炎是肿瘤患者治疗中常见的一种反应之一,严重影响到肿瘤患者治疗进程。本研究探讨Urgo水凝胶治疗Wistar大鼠Ⅱ度放射性皮炎愈合过程中创面愈合率及其机制,为临床治疗放射性皮炎提供新方法。方法建立Wistar大鼠Ⅱ度放射性皮炎模型,随机分成3组,实验组给予Ugro水凝胶0.2mm涂层,比亚芬组(传统组)给予比亚芬0.2mm涂层,生理盐水组(对照组)给予0.9%生理盐水喷雾创面处理。通过HE染色、免疫组化及图像分析,测定各组Wistar大鼠放疗后7、14、21和28d创面肉芽组织、创面面积及创面中碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)和转化生长因子-β1(transforming growth factor-β1,TGF-β1)表达。结果 X射线放射后大鼠出现脱毛或皮肤溃疡现象,实验组创面面积在第21d为(0.900±0.070)cm2,低对照组(2.317±0.280)cm2和传统组(1.550±0.080)cm2,差异有统计学意义,F=20.4,P<0.05;实验组创面面积在第28d为(1.061±0.054)cm2,明显低于对照组(2.520±0.140)cm2和传统组(1.453±0.080)cm2,差异有统计学意义,F=68.84,P<0.05。实验组第28dbFGF阳性表达量为55.00±1.00,高于传统组43.60±1.81,差异有统计学意义,t=5.52,P=0.000 6。实验组第28天TGF-β1阳性表达量为51.00±3.69,高于传统组36.40±4.41,差异有统计学意义,t=2.54,P=0.035。结论 Ugro水凝胶能加快Wistar大鼠Ⅱ度放射性皮炎创面的愈合,减轻创面的炎症反应,促进弹力纤维和胶原的修复,其机制与表皮生长因子的表达有关。 OBJECTIVE To investigate the effect of Urgo Hydrogel on theⅡ degree radiation dermatitis in Wistar rat and its influence on the histomorphological and collagenous change of wound.METHODS Thirty-six Wistar female rats withⅡdegree radiation dermatitis induced by X-RAD 225 X-ray 40 Gy one time radiation were randomly divided into3 groups and treated with Urgo Hydrogel(Experimental group),Trolamine Cream(Control group),and normal saline respectively(Normal group).The area of ulcer of all groups were measured,stained by HE staining and IHC staining for observing the changes in histomorphology and levels of bFGF bFGF,TGF-β1 in the wound tissue.RESULTS X-irradiation induced epilation or skin ulcers in almost all rats.The wound area in Urgo Hydrogel group(0.900±0.07)cm2 was lower significantly than that in the normal group(2.317±0.280)cm2 and control group(1.550±0.080)cm2 at 21 st day(F=20.4,P〈0.05).The wound area in Urgo Hydrogel group(1.061±0.0540)cm2 was lower significantly than that in the normal group(2.520±0.140)cm2 and control group(1.453±0.080)cm2 at the 28 th day(F=68.84,P〈0.05).The total expression level of bFGF in experimental group was 55.00±1.00,which was significantly higher than that in control group(43.60±1.81)at the 28 th day(t=5.52,P〈0.001).The total expression level of TGF-β1 in experimental group was 51.00±3.69,which was significantly higher than that in control group(36.40±4.41)at 28 th day(t=2.54,P=0.035).CONCLUSION The Urgo Hydrogel can promote the ulcer healing ofⅡ degree radiation dermatitis,reduce the inflammatory reaction in wound,and promote the repair of elastic fibers and collagen related with the expressions of epidermal growth factors.
